番茄黄化曲叶病毒(tomato yellow leaf curl virus,TYLCV)是一种全球性分布的植物病毒,严重为害番茄、烟草等作物.为探究其致病机制,本研究以TYLCV编码的运动蛋白V2为诱饵,从本氏烟中筛选并鉴定出与其互作的肌球蛋白NbMyosin Ⅺ-2(GenBank:DQ875135.1).通过双分子荧光互补、共定位及免疫共沉淀试验证实:V2与NbMyosin Ⅺ-2直接互作,且二者在微丝上形成点状复合体.基因沉默试验表明,沉默NbMyosin Ⅺ-2会抑制TYLCV对本氏烟的侵染,阻碍TYLCV运动复合物V2-C5向胞间连丝的定位,导致其滞留于细胞质,但不影响V2的沉默抑制子功能.此外,瞬时过表达NbMyosin Ⅺ-2显性负突变体的植株可显著抑制TYLCV、烟草曲茎病毒及云南番茄曲叶病毒的侵染.本研究揭示了NbMyosin Ⅺ-2通过与V2互作调控双生病毒的细胞间运动的分子机制,为抗病毒策略的研发提供了新靶点.

Tomato yellow leaf curl virus(TYLCV)is a globally distributed plant virus that causes severe damage to crops such as tomato and tobacco plants.To investigate its pathogenic mechanisms,the TYLCV-encoded movement protein V2 was used as bait to screen and identify interacting protein from Nicotiana benthamiana.A myosin motor protein,NbMyosin Ⅺ-2(GenBank:DQ875135.1),was identified as a V2-interacting partner.Bimolecular fluorescence complementation,co-localization,and co-immunoprecipitation assays confirmed that V2 directly interacts with NbMyosin Ⅺ-2,and that they form punctate complexes along actin filaments.Gene silencing assays revealed that knockdown of NbMyosin Ⅺ-2 inhibited TYLCV infection in N.benthamiana and disrupted the localization of the TYLCV movement complex V2-C5 to plasmodesmata,trapping it in the cytoplasm,without affecting the silencing suppressor activity of V2.Furthermore,transient overexpression of a dominant-negative mutant of NbMyosin Ⅺ-2 significantly inhibited infection by TYLCV,tobacco curly shoot virus,and tomato leaf curl Yunnan virus.This study indicates that NbMyosin Ⅺ-2 regulates the intercellular movement of geminiviruses through interaction with V2,providing a potential target for the development of antiviral strategies.
