金纳米颗粒辅助超声疗法重塑三阴性乳腺癌免疫微环境的研究进展OA
Advances in Nanoparticle-Assisted Ultrasound Therapy for Remodeling Immune Microenvironment in Triple-Negative Breast Cancer
金纳米颗粒辅助超声疗法(NAUT)通过低频超声原位激活肿瘤部位富集的金纳米颗粒,利用声空化及声动力效应产生活性氧,诱导免疫原性细胞死亡,从而将免疫抑制的"冷"型三阴性乳腺癌微环境逆转为免疫激活的"热"型.该技术凭借其物理靶向精准、微创及免疫增效等优势,在克服肿瘤异质性和清除微小残留病灶方面展现出超越传统化疗或单药免疫检查点抑制剂的潜力.本文系统综述NAUT的作用机制,重点阐述智能纳米平台的设计策略,评析其与免疫检查点抑制剂的协同疗效,并探讨其在清除微小残留病灶中的临床转化前景与挑战,旨在为三阴性乳腺癌的精准物理-免疫治疗提供新范式.
Nanoparticle-assisted ultrasound therapy(NAUT)employs low-frequency ultrasound to in situ activate gold nanoparticles,generating cavitation-and sonodynamic-derived reactive oxygen species that elicit immunogenic cell death and convert the immunologically"cold"triple-negative breast cancer(TNBC)microenvironment into a"hot"phenotype.Benefiting from its physical targeting precision,minimal invasiveness,and potent immuno-sensitization,NAUT surpasses conventional chemotherapy or single-agent immune checkpoint inhibitors in overcoming TNBC heterogeneity and eradicating minimal residual disease.In this review,the mechanistic advances of NAUT are summarized,the design of smart nanoplatforms is highlighted,its synergistic efficacy with immune checkpoint inhibitors is evaluated,and the clinical potential and challenges of NAUT in eradicating minimal residual disease are discussed.This review offers a new paradigm for precision physical-immunotherapy of TNBC.
