膜荚黄芪褐化愈伤组织的结构及代谢组分析OA
Structural and metabolomic analyses of browned callus in Astragalus membranaceus
为探究膜荚黄芪(Astragalus membranaceus)愈伤组织褐化的内在机制,以膜荚黄芪绿色愈伤组织和褐化愈伤组织为材料,采用广泛靶向代谢组学技术,结合细胞形态及超微结构观察,系统分析愈伤组织褐化过程中的形态结构和代谢物变化.结果表明,膜荚黄芪愈伤组织的褐化是由激素介导的活性氧(reactive oxygen species,ROS)过量累积所引发的,ROS通过诱导细胞膜脂质过氧化,导致叶绿体、线粒体及细胞核等超微结构破坏,并引发蛋白质及核酸的氧化损伤,最终触发愈伤组织衰老与死亡.本研究初步揭示了膜荚黄芪愈伤组织褐化的可能原因,为解决组织培养技术瓶颈、推动优质种苗规模化生产提供理论依据.
To investigate the underlying mechanism of callus browning in Astragalus membranaceus,green callus and browned callus were used as materials.Widely targeted metabolomics,combined with cellular morphology and ultrastructure observations,was employed to systematically analyze the morphological and metabolic changes during browning.The results showed that the browning process was initiated by hormone-mediated excessive accumulation of reactive oxygen species(ROS).The ROS induced lipid peroxidation in cell membranes,which led to the disintegration of ultrastructures such as chloroplasts,mitochondria,and nuclei,and also caused oxidative damage to proteins and nucleic acids.Ultimately,these effects triggered callus senescence and death.In conclusion,this study indicates that callus browning in A.membranaceus is a consequence of ROS overaccumulation,which drives a cascade of cellular damage culminating in death.These findings elucidate the potential mechanisms underlying this phenomenon and provide a theoretical foundation for overcoming key bottlenecks in tissue culture,thereby facilitating the large-scale production of high-quality plantlets.
