阻断Wee1激酶诱导有丝分裂灾难以抑制猪圆环病毒2型复制OA
Wee1 inhibition induces mitotic catastrophe to suppress porcine circovirus type 2 replication
猪圆环病毒2型(porcine circovirus type 2,PCV2)是猪圆环病毒相关疾病的主要病原体,严重危害全球养猪业.本研究旨在评估靶向宿主细胞周期调控因子Wee1激酶对PCV2复制的影响.在体外感染模型中,结合染色体铺展、间接免疫荧光检测及蛋白质印迹法,系统分析PCV2感染对宿主细胞DNA损伤应答(DNA damage response,DDR)及细胞周期进程的影响,并评估Wee1抑制剂MK-1775的抗病毒作用.结果表明,PCV2感染可显著激活宿主DDR,84%的感染细胞呈γ-H2AX阳性,并导致细胞周期阻滞于S期.进一步研究发现,MK-1775处理可诱导约60%的感染细胞发生有丝分裂灾难,同时显著降低PCV2感染率及病毒蛋白表达水平.以上结果表明,PCV2复制依赖宿主细胞周期调控,而Wee1抑制可通过破坏细胞周期检查点并诱导有丝分裂灾难,有效抑制病毒复制.因此,靶向宿主细胞周期调控因子Wee1可能成为干预PCV2感染的潜在策略.
Porcine circovirus type 2(PCV2)is the primary pathogen of porcine circovirus-associated diseases,posing a serious threat to the global swine industry.This study aimed to evaluate the effect of targeting the host cell cycle regulator Wee1 kinase on PCV2 replication.Using an in vitro infection model,we systematically analyzed the impact of PCV2 infection on the host DNA damage response(DDR)and cell cycle progression through a chromosome spread assay,indirect immunofluorescence assay,and Western blotting analyses,and assessed the antiviral activity of the Wee1 inhibitor MK-1775.The results showed that PCV2 infection significantly activated host DDR,with 84%of infected cells exhibiting γ-H2AX positivity,and induced cell cycle arrest in the S phase.Further analysis revealed that MK-1775 treatment triggered mitotic catastrophe in approximately 60%of infected cells,accompanied by a significant reduction in the PCV2 infection rate and viral protein expression levels.These findings indicate that PCV2 replication relies on host cell cycle regulation,and that Wee1 inhibition effectively suppresses viral replication by disrupting cell cycle checkpoints and inducing mitotic catastrophe.Thus,targeting the host cell cycle regulator Wee1 may represent a potential strategy for intervening in PCV2 infection.
