Research progress on calpain 3-mediated intranuclear proteolytic pathways

钙蛋白酶3(calpain 3,CAPN3)是钙蛋白酶家族中唯一含有核定位信号的成员.CAPN3最先在骨骼肌中发现,并曾被认为具有组织特异性,但近年研究表明,其在包括肝脏在内的多种组织中广泛表达,参与调控底物活性、稳定性及亚细胞定位等过程.随着CAPN3核内分布的确认,其介导的核内蛋白水解途径逐渐受到关注.本文综述了CAPN3介导的核内蛋白水解途径的最新进展,重点阐述了核仁消化器官延伸因子(digestive-organ expansion factor,DEF)-CAPN3通路在维持核仁蛋白稳态中的作用,以及新鉴定的CAPN3-多梳抑制复合物1(polycomb repressive complex 1,PRC1)通路如何通过蛋白酶解影响表观遗传调控,并对未来的研究方向进行了展望.

Calpain 3(CAPN3)is the sole member of the calpain family that possesses a nuclear localization signal.Although it was initially identified in skeletal muscle and was long regarded as tissue specific,recent studies have demonstrated that CAPN3 is widely expressed across multiple tissues,including the liver,and plays important roles in regulating substrate activity,stability,and subcellular localization.The confirmation of its nuclear presence has heightened interest in CAPN3-mediated intranuclear proteolytic pathways.This review summarizes recent advancements in CAPN3-mediated intranuclear proteolytic pathways,with a particular focus on the nucleolar digestive-organ expansion factor(DEF)-CAPN3 pathway,which is essential for maintaining nucleolar protein homeostasis,and the newly identified CAPN3-polycomb repressive complex 1(PRC1)pathway,which connects proteolysis with epigenetic regulation.Finally,potential avenues for future research are discussed.
