向日葵不同器官响应列当寄生的转录组分析OA
Transcriptome Analysis of Different Organs in Helianthus annuus in Response to Orobanche Parasitism
向日葵列当寄生危害呈逐年加重趋势,已严重制约向日葵产量与品质提升.为解析向日葵响应列当寄生的分子调控机制及代谢通路,以当地主栽品种SH361为试验材料,设置列当侵染组与正常生长对照组,采用转录组测序(RNA-seq)结合生物信息学分析方法,对根、茎、叶、花瓣、种子等5个器官的基因表达模式进行探究.结果显示,试验共获得219.24 Gb Clean Data,GC含量为44.34%~45.47%,Q20、Q30碱基占比分别不低于98.06%、94.10%,测序数据质量优良.对比分析分别鉴定出差异表达基因1 531、2 314、2 290、2 137、1 658条,其中根部差异基因数量最少,茎部最多.对差异表达基因进行KEGG通路富集分析发现,MAPK信号通路-植物是其核心富集通路,且不同器官的通路富集特征存在明显差异.这些差异基因通过调控mRNA转录及抗性激素合成代谢,协同介导向日葵对列当寄生的防御响应.
The damage caused by Orobanche parasitism on Helianthus annuus has been increasing year by year,severely restricting the improvement of sunflower yield and quality.To elucidate the molecu-lar regulatory mechanisms and metabolic pathways underlying the response of Helianthus annuus to Orobanche parasitism,the locally dominant cultivar SH361 was used as the experimental material.An Orobanche-infected group and a normal growth control group were established,and transcriptome se-quencing(RNA-seq)combined with bioinformatics analysis was employed to investigate the gene ex-pression patterns in 5 organs:roots,stems,leaves,petals,and seeds.The results showed that a total of 219.24 Gb of clean data was obtained,with GC contents ranging from 44.34%to 45.47%,and Q20 and Q30 base percentages not lower than 98.06%and 94.10%,respectively,indicating high sequen-cing data quality.Comparative analysis identified 1 531,2 314,2 290,2 137,and 1 658 differentially expressed genes(DEGs)in the respective organs,with the fewest DEGs in roots and the most in stems.KEGG pathway enrichment analysis of the DEGs revealed that the MAPK signaling pathway-plant was the core enriched pathway,and the pathway enrichment characteristics differed markedly a-mong organs.These DEGs synergistically mediate the defense response of Helianthus annuus to Oro-banche parasitism by regulating mRNA transcription and the biosynthesis and metabolism of resist-ance-related hormones.
