不同类型钠盐胁迫对玉米自交系B73萌发的影响OA
The Effects of Different Types of Sodium Salt Stress on Germination of Maize Inbred Line B73
萌发期是玉米响应盐胁迫最敏感阶段,解析其耐盐机理对玉米抗逆育种具有重要意义.以玉米自交系B73为材料,分析不同浓度中性盐(NaCl、Na2SO4)和碱性盐(NaHCO3、Na2CO3)处理下的萌发指标、生理特征及盐胁迫响应基因的表达变化.结果表明,玉米萌发指标随盐浓度升高显著变化,其中在150 mmol/L各种钠盐胁迫下,种子萌发受显著抑制,Na2CO3抑制效应最突出,发芽势、发芽率和发芽指数分别降至ck的86.3%、81.8%和84.7%,盐害指数升高至85%;胚根与胚芽中抗氧化酶活性呈明显组织特异性和盐分依赖性,胚根中SOD、POD、CAT、APX活性受显著抑制,碱性盐胁迫最为突出,综合得出碱性盐胁迫对植物的伤害显著强于中性盐,且不同类型钠盐在150 mmol/L胁迫下与ck差异显著;盐胁迫相关基因表达分析发现,ZmSOD3在NaCl和NaHCO3胁迫下极显著上调,ZmPOD、ZmPOD44、ZmPRX1和ZmCAT1在NaHCO3胁迫下极显著上调,ZmGT61和ZmMAPK4在Na2CO3胁迫下显著上调,ZmMAC39和ZmMAC82在盐处理下显著下调.综上,钠盐胁迫对玉米种子萌发的抑制呈浓度依赖性,Na2CO3胁迫效应最显著,不同钠盐胁迫存在基因表达特异性调控.
The germination stage is the most sensitive phase of maize(Zea mays L.)in response to salt stress,and elucidating its salt tolerance mechanisms is of great significance for stress-resistant breed-ing in maize.Using the maize inbred line B73 as the material,the germination indicators,physiologi-cal characteristics,and expression changes of salt stress-responsive genes were analyzed under treat-ments with different concentrations of neutral salts(NaCl,Na2SO4)and alkaline salts(NaHCO3,Na2CO3).The results showed that the germination indicators of maize changed significantly with in-creasing salt concentrations.Seed germination was markedly inhibited under 150 mmol/L stress for all sodium salts,with Na2CO3 exhibiting the most prominent inhibitory effect.The germination vigor,germination percentage,and germination index decreased to 86.3%,81.8%,and 84.7%of the con-trol,respectively,and the salt injury index increased to 85%.The activities of antioxidant enzymes in the radicle and plumule displayed distinct tissue specificity and salt dependency;the activities of SOD,POD,CAT,and APX in the radicle were significantly inhibited,with alkaline salt stress being the most pronounced.Overall,alkaline salt stress caused significantly greater damage to plants than neu-tral salt stress,and different types of sodium salts at 150 mmol/L showed significant differences com-pared with the control.Expression analysis of salt stress-related genes revealed that ZmSOD3 was highly significantly upregulated under NaCl and NaHCO3 stress,ZmPOD,ZmPOD44,ZmPRX1,and ZmCAT1 were highly significantly upregulated under NaHCO3 stress,ZmGT61 WTBZ and ZmMAPK4 were significantly upregulated under Na2CO3 stress,and ZmMAC39 and ZmMAC82 were significantly downregulated under salt treatments.In conclusion,the inhibition of maize seed germination by sodium salt stress was concentration-dependent,with Na2CO3 stress exhibiting the most significant effect,and gene expression regulation was specific to different sodium salt stresses.
