SAP proteins,characterized by the A20/AN1 zinc finger domain,constitute a class of stress-associated proteins,and their family members play crucial roles in the abiotic stress responses of plants.Bioinformatic methods were employed to identify the SAP gene family members in the Chrys-anthemum indicum genome,and analyses of gene structure,conserved motifs,and physicochemical properties were conducted.A total of 14 CnSAP gene family members were identified,most of which encode proteins containing typical N-terminal A20 and C-terminal AN1 domains,although a few members exhibited domain deletions.Conserved motif analysis revealed that Motif1 and Motif2 corre-sponded to the AN1 and A20 domains,respectively,indicating that the overall structure of the family is highly conserved.Phylogenetic analysis classified the CnSAP,AtSAP,and OsSAP proteins into five subfamilies,with some CnSAP members clustering in the same clade as known stress-related SAP genes from Arabidopsis thaliana or Oryza sativa.Physicochemical property predictions showed that all CnSAP proteins are hydrophilic basic proteins,with random coils dominating their secondary structures.Subcellular localization predictions indicated that CnSAP proteins are primarily distributed in the nucleus,chloroplasts,and cytoplasm,with CnSAP14 specifically localized to the chloroplast.Analysis of cis-acting elements in the promoter regions revealed that all CnSAP members are enriched in low-temperature-responsive elements,light-responsive elements,and various hormone-responsive elements.Expression analysis under low-temperature stress demonstrated that the CnSAP family members exhibit diverse expression patterns,suggesting that different members play differentiated functions in the cold response regulatory network of Chrysanthemum indicum.
