为探究人参成花相关差异基因及其内源激素变化,挖掘成花关键因子,以3个品系人参开花与不开花的植株叶片为材料,采用高通量转录组测序技术进行转录组比较分析,结合液相色谱-串联质谱技术测定内源激素含量,并利用qRT-PCR对关键基因进行表达验证,共获得63 819个基因,筛选出成花相关差异表达基因2 365个.GO和KEGG富集分析显示,差异基因显著富集于次生代谢产物合成与植物激素信号转导等通路.WGCNA分析筛选出与赤霉素、细胞分裂素显著正相关的blue核心模块,PPI网络分析鉴定出LFY、HTR2、F24B9.25为成花调控网络的核心枢纽基因.qRT-PCR验证结果与转录组测序一致.明确了 LFY、HTR2、F24B9.25等基因在人参开花植株中上调表达,揭示了赤霉素与细胞分裂素信号途径在人参成花调控中的潜在关键作用.

To investigate the differentially expressed genes(DEGs)and endogenous hormone changes associated with flowering in Panax ginseng and to identify key regulatory factors,leaves from flow-ering and non-flowering plants of 3 Panax ginseng lines were used as materials.High-throughput transcriptome sequencing was employed for comparative transcriptomic analysis,endogenous hor-mone contents were determined using liquid chromatography-tandem mass spectrometry(LC-MS/MS),and the expression of key genes was validated by qRT-PCR.A total of 63 819 genes were ob-tained,and 2 365 DEGs related to flowering were identified.GO and KEGG enrichment analyses showed that the DEGs were significantly enriched in pathways such as secondary metabolite biosyn-thesis and plant hormone signal transduction.WGCNA identified a blue core module that was signifi-cantly positively correlated with gibberellin and cytokinin.PPI network analysis identified LFY,HTR2,and F24B9.25 as core hub genes in the flowering regulatory network.The qRT-PCR valida-tion results were consistent with the transcriptome sequencing data.This study confirmed that genes such as LFY,HTR2,and F24B9.25 were upregulated in flowering plants of Panax ginseng,revea-ling the potential key roles of gibberellin and cytokinin signaling pathways in the regulation of flower-ing in Panax ginseng.
