为建立藏药材西藏千里光(Senecio tibeticus Hook.f.)快速繁育体系,分别探究了西藏千里光种子萌发与幼苗组织培养.研究不同消毒方式处理西藏千里光种子,并结合不同浓度(1%、2%、3%)H2O2浸种后在不同培养基观察种子萌发情况.结果表明,西藏千里光种子以0.1%HgCl2溶液消毒5 min后灭菌更彻底;种子萌发室内的平均发芽率为81.6%;室外的平均发芽率为57.3%;较高浓度(2%~3%)H2O2在MS培养基处理组呈现出最佳的生长状态,而1%H2O2在含1 mg/L 6-BA+1 mg/L NAA的培养基中对种子萌发的促进效果最佳.以西藏千里光幼苗根系为材料进行组织培养研究,配制诱导愈伤组织、诱导丛芽和诱导生根培养基,各阶段中观察生长情况.成功诱导愈伤组织,实现丛芽分化3个,生根诱导培养每株生长2~5条.平均生根率为70%;西藏千里光幼苗根系从愈伤组织成功长出丛芽并生根.

To establish a rapid propagation system for the Tibetan medicinal plant Senecio tibeticus,seed germination and seedling tissue culture of Senecio tibeticus were investigated separately.Seeds of Senecio tibeticus were treated with different disinfection methods,followed by soaking in different concentrations(1%,2%,3%)of H2O2,and then cultured on different media to observe seed germi-nation.The results showed that seeds of Senecio tibeticus were most effectively sterilized by treatment with 0.1%HgCl2 solution for 5 min.The average germination percentage in the indoor germination chamber was 81.6%,while the outdoor average was 57.3%,Higher concentrations(2%-3%)of H2O2 on MS medium showed the best growth performance,whereas 1%H2O2 on medium containing 1 mg/L 6-BA+1 mg/L NAA exhibited the best promoting effect on seed germination.Using the roots of Senecio tibeticus seedlings as materials for tissue culture,media for callus induction,shoot cluster induction,and rooting induction were prepared,and growth was observed at each stage.Callus was successfully induced,three shoot clusters were differentiated,and each plantlet developed 2-5 roots during rooting induction.The average rooting rate was 70%.Shoot clusters were successfully regenerated from callus derived from the seedling roots of Senecio tibeticus and subsequently rooted.
