目的 基于网络药理学和动物实验探究黄芩汤通过调控PI3K/AKT/NF-κB信号通路缓解溃疡性结肠炎(UC)的作用机制,为其临床应用提供依据.方法 采用网络药理学预测黄芩汤缓解UC的有效成分及潜在靶点,筛选核心成分、靶点及通路,通过分子对接验证成分与靶点的结合能力.以葡聚糖硫酸钠构建小鼠UC模型,将小鼠分为正常组、模型组、黄芩汤组,每组7只,并予相应干预7 d.监测小鼠体质量,进行疾病活动指数(DAI)评分,观察结肠形态并测量长度,HE染色观察结肠组织形态并进行病理评分,ELISA检测血清促炎因子肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β及IL-6含量,Western blot检测结肠组织相关通路蛋白表达.结果 网络药理学筛选得到黄芩汤缓解UC的核心有效成分4个、核心靶点10个,分子对接显示成分与靶点结合稳定.动物实验表明,黄芩汤可显著降低模型小鼠DAI评分(P<0.01),恢复结肠长度(P<0.01),减轻结肠病理损伤(P<0.01),下调血清IL-1β、IL-6、TNF-α含量(P<0.01),抑制结肠组织p-AKT、p-NF-κB p65蛋白表达(P<0.01,P<0.05).结论 黄芩汤可通过抑制PI3K/AKT/NF-κB信号通路异常激活,减少炎症因子释放,减轻肠道炎症损伤,从而缓解UC.

Objective To explore the mechanism of Huangqin Decoction in alleviating ulcerative colitis(UC)by regulating the PI3K/AKT/NF-κB signaling pathway based on network pharmacology and animal experiment;To provide basis for its clinical application.Methods Network pharmacology was used to predict the effective components and potential targets of Huangqin Decoction in alleviating UC,and screen core components,targets,and pathways.The binding ability between components and targets was validated through molecular docking.A mouse UC model was constructed using dextran sulfate sodium.The mice were divided into normal group,model group,and Huangqin Decoction group,with 7 mice in each group,and were given corresponding intervention for 7 d.The body mass of mice was monitored,Disease Activity Index(DAI)scoring was performed,colon morphology was observed,and length was measured.HE staining was used to observe the morphology of colon tissue and perform histopathological scoring.ELISA was used to detect the contents of serum pro-inflammatory factors TNF-α,IL-1β and IL-6.Western blot was used to detect the expressions of related pathway proteins in colon tissue.Results Four core components and ten core targets of Huangqin Decoction in alleviating UC were screened,with stable binding verified by molecular docking.Animal experiments showed that Huangqin Decoction could significantly reduce DAI score(P<0.01),restore colon length(P<0.01),alleviate colonic pathological damage(P<0.01),down-regulate serum IL-1β,IL-6 and TNF-α contents(P<0.01),and inhibit p-AKT and p-NF-κB p65 protein expressions in colon tissue(P<0.01,P<0.05).Conclusion Huangqin Decoction can alleviate UC by inhibiting the abnormal activation of PI3K/AKT/NF-κB pathway,reducing inflammatory factor release and intestinal inflammatory injury.
