痰热清注射液雾化吸入对慢性支气管炎急性发作大鼠SP、NEP表达影响的实验研究OA
Experimental Study on Nebulized Inhalation of Tanreqing Injection(痰热清注射液)on Sp and Nep Expressions in Rats With Acute Exacerbation of Chronic Bronchitis
目的:探讨痰热清注射液雾化吸入对慢性支气管炎急性发作大鼠SP、NEP表达的影响.方法:60 只SD大鼠随机分为正常对照组、模型组、沐舒坦组、痰热清低、中、高剂量组,每组10 只.采用小剂量脂多糖(LPS)诱导模拟革兰阴性菌感染联合被动吸烟法造模.ELISA法检测血清和肺泡灌洗液(BALF)中速激肽P物质(SP)和中性内肽酶(NEP)含量,HE染色法观察肺组织病理表现,免疫组化检测肺组织SP和NEP蛋白表达并进行半定量评分.结果:与模型组比较,沐舒坦组及痰热清低、中、高剂量组血清和BALF中SP明显降低,NEP明显升高,差异有统计学意义(P<0.05).肺组织 SP 和NEP免疫组化检测结果显示,与模型组比较,痰热清各组和沐舒坦组肺组织中SP蛋白表达水平均降低,NEP蛋白表达水平均升高,差异有统计学意义(P<0.05);与沐舒坦组比较,痰热清高剂量组SP、NEP蛋白表达比较,差异有统计学意义(P<0.05).结论:痰热清注射液雾化吸入可以有效降低血清和BALF中SP含量及肺组织中蛋白的表达,增加血清和BALF中NEP含量及肺组织中蛋白表达,减轻气道神经源性炎症.痰热清高剂量组疗效优于沐舒坦,且痰热清各组剂量与疗效之间存在正相关的量效关系.
Objective:To investigate the effects of aerosol inhalation of Tanreqing injection on the expression of SP and NEP in rats with acute exacerbation of chronic bronchitis.Methods:60 SD rats were randomly divided into normal control group,model group,Ambroxol group,and low-,medium-and high-dose Tanreqing injection groups,with 10 rats in each group.The rat model of acute exacerbation of chronic bronchitis was established by low-dose lipopolysaccharide(LPS)induction to simulate Gram-negative bacterial infection combined with passive smoking method.The contents of substance P(SP)and neutral endopeptidase(NEP)in serum and bronchoalveolar lavage fluid(BALF)were determined by ELISA.HE staining was used to observe the pathological changes of lung tissue.Immunohistochemistry was applied to detect the protein expressions of SP and NEP in lung tissue,followed by semi-quantitative scoring.Results:Compared with the model group,the levels of SP were significantly decreased,while the levels of NEP were significantly increased in serum and BALF of the ambroxol group and all Tanreqing injection treatment groups(P<0.05).Immunohistochemical results showed that,compared with the model group,the protein expression of SP was downregulated and NEP was upregulated in the lung tissues of each treatment group,with statistically significant differences(P<0.05).In addition,the high-dose Tanreqing Injection group exhibited significant differences in SP and NEP protein expression compared with the ambroxol group(P<0.05).Conclusion:Aerosol inhalation of Tanreqing Injection can effectively reduce the levels of SP in serum and BALF,as well as the protein expression of SP in lung tissue,while increase the contents of NEP in serum,BALF and lung tissue.This regulatory effect helps alleriate airway neurogenic inflammation.
