目的:探讨地黄饮子对脑缺血再灌注损伤(cerebral ischema-reperfusion injury,CIRI)的改善作用及其分子机制.方法:构建CIRI大鼠模型,将30 只SD大鼠随机分为假手术组、模型组、地黄饮子低剂量组(6 g/kg)、地黄饮子高剂量组(12 g/kg)、阳性对照组(尼莫地平,10 mg/kg),每组6 只,灌胃给药,每日1 次,连续给药28 d.使用改良神经系统严重程度评分(modified neurological severity scores,mNSS)评估大鼠的神经功能损伤,HE 染色法观察脑组织的病理变化,ELISA 法检测脑组织中的 TNF-α、IL-6、IL-1β、MDA、SOD和GSH-PX水平.透射电镜和Western blot法观察脑组织自噬体和线粒体形态变化及LC3II/LC3I、Beclin1、Fis1、Drp1 蛋白表达.结果:与模型组比较,地黄饮子低、高剂量组和尼莫地平组能减少大鼠mNSS评分,改善脑病理损伤,降低TNF-α、IL-6、IL-1β、MDA水平,升高SOD、GSH-PX水平;可减少自噬体数量,增加线粒体数量,并降低LC3II/LC3I、Beclin1、Fis1、Drp1 蛋白表达.结论:地黄饮子对CIRI具有改善作用,这可能与抑制线粒体分裂和自噬过程有关.

Objective:To explore the therapeutic effect of Dihuang Yinzi decoction on cerebral ischemia-reperfusion injury(CIRI)and its underlying molecular mechanism.Methods:A rat model of cerebral ischemia-reperfusion injury(CIRI)was established.30 SD rats were randomly divided into five groups,with 6 rats per group:sham operation group,model group,low-dose Dihuang Yinzi group(6 g/kg),high-dose Dihuang Yinzi group(12 g/kg),and positive control group(nimodipine,10 mg/kg).All rats were administered intragastrically once a day for 28 consecutive days.Modified neurological severity scores(mNSS)were used to assess neurological dysfunction in rats.Hematoxylin-eosin(HE)staining was applied to observe the pathological changes of brain tissue.The levels of TNF-α,IL-6,IL-1β,MDA,SOD and GSH-PX in brain tissue were detected by ELISA.Transmission electron microscopy(TEM)and Western blot were adopted to observe the morphological changes of autophagosomes and mitochondria in brain tissue,as well as the protein expressions of LC3II/LC3I,Beclin1,Fis1 and Drp1.Results:Compared with the model group,both low-and high-dose Dihuang Yinzi groups and the nimodipine group significantly decreased the mNSS scores and alleviated cerebral pathological damage in rats.Meanwhile,the levels of proinflammatory factors(TNF-α,IL-6,IL-1β)and oxidative stress indicator MDA were markedly reduced,while the activities of antioxidant enzymes SOD and GSH-PX were notably elevated.In addition,Dihuang Yinzi and nimodipine treatment reduced the number of autophagosomes,increased the number of mitochondria,and downregulated the protein expression levels of LC3II/LC3I,Beclin1,Fis1 and Drp1 in brain tissue.Conclusion:Dihuang Yinzi decoction exerts a protective effect against cerebral ischemia-reperfusion injury,which may be closely related to the inhibition of mitochondrial fission and excessive autophagy.
