非小细胞肺癌组织中Wilms肿瘤1相关蛋白、含PEST的核蛋白表达与病理特征及预后的关系OA
The relationship between the expression of Wilms tumor 1-associated protein and PEST-containing nuclear protein in non-small cell lung cancer and pathological features and prognosis
目的 探讨非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中 Wilms肿瘤1相关蛋白(Wilms tumor 1-associated protein,WTAP)、含PEST的核蛋白(PEST-containing nuclear protein,PCNP)的表达特点,分析其与NSCLC病理特征以及预后的关系.方法 选取2018年2月至2020年2月许昌市中心医院收治的189例NSCLC患者,收集癌组织及配对癌旁组织标本,应用免疫组织化学检测组织中 WTAP、PCNP表达情况,比较不同病理特征NSCLC组织中 WTAP、PCNP表达差异.术后对患者进行为期5年的随访,利用 Kaplan-Meier生存曲线比较不同 WTAP、PCNP表达水平患者的OS和PFS差异,并采用多因素Cox比例风险回归模型评估 NSCLC患者预后的危险因素.结果 NSCLC癌组织 WTAP、PCNP阳性率高于癌旁组织(P<0.05),且 WTAP与PCNP表达呈正相关(rs=0.502,P<0.001).TNM 分期Ⅲa期、低分化、淋巴结转移、肿瘤浸润的NSCLC组织中 WTAP、PCNP阳性率均高于TNM 分期Ⅰ～Ⅱ期、中高分化、无淋巴结转移、无肿瘤浸润的 NSCLC组织(均P<0.05).随访期间失访3例,复发25例,转移17例,死亡59例,存活127例.WTAP、PCNP阳性表 达 NSCLC 患者5年无进展生存率、5年总生存率低于 WTAP、PCNP 阴性表达 NSCLC 患者(WTAP分别为37.89%比53.85%,63.64%比79.63%,Log-rank χ2=12.890、4.720,均P<0.05;PCNP 40.68%比54.41%,62.71%比77.94%,Log-Rank χ2=5.122、5.131,均P<0.05).多因素Cox比例风险回归模型显示,TNM 分期Ⅲa期、WTAP阳性表达、PCNP阳性表达是 NSCLC患者随访期间死亡的危险因素(P<0.05).结论 NSCLC组织中WTAP、PCNP阳性表达与恶性病理特征以及不良预后有关.
Objective To investigate the expression characteristics of Wilms tumor 1-associated protein(WTAP)and PEST-containing nuclear protein(PCNP)in non-small cell lung cancer(NSCLC)tissues and to analyze their correlations with pathological features and prognosis.Methods From February 2018 to February 2020,189 NSCLC patients admitted to Xuchang Central Hospital were enrolled.Cancer tissues and paired adjacent non-tumor tissues were enrolled.Cancer tissue specimens and paired adjacent non-cancerous tissue specimens were collected.The expression of WTAP and PCNP in tissues was detected by immunohistochemistry.The expression differences of WTAP and PCNP in NSCLC tissues with different pathological characteristics were compared.The patients were followed up for 5 years after the operation.The Kaplan-Meier survival curve was used to compare the differences in overall survival(OS)and progressin free survival(PFS)among patients with different expression levels of WTAP and PCNP,and the multivariate Cox proportional hazards regression model was used to evaluate the risk factors affecting the prognosis of NSCLC patients.Results The positive rates of WTAP and PCNP in NSCLC cancer tissues were higher than those in adjacent tissues(P<0.05),and the expression of WTAP was positively correlated with that of PCNP(rs=0.502,P<0.001).The positive rates of WTAP and PCNP in NSCLC tissues with TNM stageⅢa,poor differentiation,lymph node metastasis and tumor invasion were all higher than those in NSCLC tissues with TNM stage Ⅰ-Ⅱ,moderate to high differentiation,no lymph node metastasis and no tumor invasion(P<0.05).During the follow-up period,3 cases were lost to follow-up,25 cases relapsed,17 cases metastasized,59 cases died,and 127 cases survived.The 5-year PFS rate and 5-year OS rate of NSCLC patients with positive expression of WTAP and PCNP were lower than those of NSCLC patients with negative expression of WTAP and PCNP(WTAP:37.89%vs.53.85%,63.64%vs.79.63%,Log-rank χ2=12.890,4.720,all P<0.05;PCNP:40.68%vs.54.41%,62.71%vs.77.94%,Log-rank χ2=5.122,5.131,all P<0.05).Multivariate Cox regression showed that TNM stageⅢa,positive expression of WTAP,and positive expression of PCNP were risk factors for death in NSCLC patients during the follow-up period(P<0.05).Conclusions The positive expressions of WTAP and PCNP in NSCLC tissues are related to malignant pathological characteristics and poor prognosis.
