Drug memory and synaptic plasticity in drug addiction
毒品成瘾是一种复杂的神经精神疾病,其本质是大脑奖赏系统与记忆系统的长期适应性改变导致的毒品滥用.近年来,病理性毒品记忆与神经突触可塑性的相互作用成为成瘾机制研究的核心方向.病理性毒品记忆是个体对毒品相关线索形成的高度巩固且难以消退的条件性记忆,其异常牢固与无法遗忘是复吸行为的关键驱动因素.神经突触可塑性则通过突触结构与功能的动态调整,介导毒品对神经回路的长期改造.本文综述了病理性毒品记忆与神经突触可塑性的关联机制,阐释了甲基苯丙胺、可卡因、大麻、海洛因及新精神活性物质等不同类型毒品,通过调控不同突触可塑性的特异性通路与靶点,介导毒品记忆编码、巩固与提取的具体分子和细胞机制,旨在为靶向记忆干预与突触可塑性调控的成瘾治疗策略提供理论依据与研究方向.
Drug addiction is a complex neuro-psychiatric disorder involving long-term adaptive brain changes in reward and memory that lead to drug abuse.The interaction between pathological drug memory and neural synaptic plasticity has become the core focus of addiction mechanism research.Pathological drug memory is a highly consolidated and difficult-to-erase conditional memory formed in response to drug-related cues,its abnormal firmness and inability to be forgotten are the key driving factors for relapse behavior.Neural synaptic plasticity mediates long-term neural circuitry modification by drugs through the dynamic synaptic structure and function adjustment.This article reviews the association between pathological drug memory and neural synaptic plasticity and explains the specific molecular and cellular mechanisms of drug memory encoding,consolidation,and retrieval for different types of drugs(including methamphetamine,cocaine,cannabis,heroin,and new psychoactive substances).By examining the specific pathways and targets that regulate different synaptic plasticities,this article aims to provide a theoretical basis and research directions for addiction treatment strategies targeting memory intervention and synaptic plasticity regulation.
