Selection and application of epilepsy animal models
为解决癫痫发病机制未明、药物难治性癫痫高达30%的临床难题,为癫痫研究与新药研发提供可靠工具,本研究将从癫痫动物模型分类、建模方法、模型评估三部分进行系统性梳理.模型分类上,按发作频率、年龄段、国际抗癫痫联盟(International League Against Epilepsy,ILAE)标准多角度划分模型类型并分析应用场景;建模方法上,综述体外与体内(化学法、物理法、基因编辑等)建模技术特点及优劣势;模型评估上,以行为学分析为基础,辅助脑电图(electroencephalogram,EEG)及组织学验证,进一步排除假阳性并确保模型精准有效.此外,本综述指出并总结了现阶段面临物种病理差异、耐药性模拟不足及共病模型构建困难等核心挑战.该综述为模型选择提供多方面参考依据,助力科研人员依据研究目的与实验条件精准匹配动物模型,提升研究效率,推动癫痫机制解析与临床转化研究深入发展.
This review aims to address the clinical challenges of unclear pathogenesis of epilepsy and the fact that drug-resistant epilepsy accounts for up to 30%,as well as to provide a reliable tool for epilepsy research and new drug development.Three core aspects of epilepsy animal models are summarized:classification,establishment method,and evaluation systems.Models were classified from multiple perspectives,including seizure frequency,age group,and criteria of the International League Against Epilepsy,and corresponding application scenarios were analyzed.The technical characteristics,advantages,and disadvantages of in vitro and in vivo models(including chemical,physical,and gene editing approaches)are reviewed.Behavioral analysis was found to serve as the foundation for evaluation,supplemented by electroencephalogram detection and histological verification to further exclude false positives and ensure model accuracy and effectiveness.Additionally,key challenges are identified and discussed,such as species-specific pathological differences,insufficient drug resistance simulation,and difficulties in constructing comorbidity models.The findings provide comprehensive references for model selection,assisting researchers in accurately matching animal models with their research objective and experimental conditions,improving research efficiency,and promoting the in-depth development of epilepsy mechanism research and clinical translation.
