幼年大鼠LED光源性近视模型的建立OA
Establishment of a myopia model in juvenile rats using light-emitting diodes
目的 观察和评估应用大鼠建立发光二极管(LED)光源性近视模型的方法.方法 选择 3 周龄的 SPF 级雄性大鼠 40 只,随机分为对照组、模型组,每组 20 只.在实验前和实验第 2、4、6 周对各组大鼠进行眼轴测量;采用苏木素-伊红(HE)染色法观察大鼠巩膜组织形态学变化;采用酶联免疫吸附测定法检测血清褪黑素浓度;采用 Western Blot 检测视网膜组织中基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶抑制剂-2(TIMP-2)蛋白表达,同时采用 RT-qPCR 法检测巩膜组织中二者 mRNA 表达情况.结果 与对照组相比,模型组大鼠眼轴明显延长,在第 4、6 周均有统计学意义(P<0.05),巩膜胶原纤维排列紊乱、分布稀疏且巩膜层变薄,血清褪黑素浓度显著降低(P<0.05),且视网膜组织中 MMP-2 蛋白及巩膜组织中 MMP-2 mRNA 表达均显著上调,而 TIMP-2表达则显著下调(P<0.05).结论 长期 LED 光源暴露可通过诱导眼轴延长、破坏巩膜结构完整性、调控巩膜 MMP-2/TIMP-2信号通路,成功建立了 LED 光源性近视大鼠模型.
Objective To establish and evaluate a rat model of myopia induced by light-emitting diode(LED)light exposure.Methods Forty 3-week-old male SPF rats were randomly divided into control and model groups(n=20).Axial length was measured before the experiment and at weeks 2,4,and 6.Scleral morphological changes were observed using hematoxylin and eosin(HE)staining.Serum melatonin concentration was measured by enzyme-linked immunosorbent assay.Western Blot was applied to determine matrix metalloproteinase-2(MMP-2)and tissue inhibitor of metalloproteinases-2(TIMP-2)protein expression in retinal tissue,and RT-qPCR was used to detect their messenger RNA(mRNA)expression in scleral tissue.Results The model group showed significantly elongated axial lengths at weeks 4 and 6 compared with the control group(P<0.05).Scleral tissue exhibited disorganized and sparse collagen fibers,scleral layer thinning,and a significantly reduced serum melatonin concentration(P<0.05).In addition,MMP-2 protein expression in retinal tissue and mRNA expression in scleral tissue were significantly upregulated,while TIMP-2 expression was significantly downregulated(P<0.05).Conclusions Long-term LED light exposure can induce axial elongation,destroy scleral structural integrity,and regulate scleral MMP-2/TIMP-2 signaling,successfully establishing a rat model of LED light-induced myopia.
