甘肃某规模化奶牛场群发性犊牛腹泻案例分析OA
Case Analysis of an Outbreak of Calf Diarrhea at a Large-Scale Dairy Farm in Gansu
为明确甘肃省某规模化奶牛场犊牛腹泻的病因,并提供可行的减抗防治方案,采集该牛场19头腹泻犊牛的粪便通过RT-qPCR法、饱和盐水漂浮法对腹泻病原进行检测,采集犊牛的饮用水进行pH和细菌菌落总数测定以及对细菌进行分离鉴定,采集饲料通过ELISA法进行毒素检测.结果显示,产气荚膜梭菌(Clostridium perfringens,Cp)核酸阳性率为52.63%(10/19),隐孢子虫(Cryptosporidium,COWP)核酸阳性率为5.26%(1/19),未检出牛轮状病毒(Bovine Rotavirus,BRV)、牛冠状病毒(Bovine Coronavirus,BCoV)、牛病毒性腹泻病毒(Bovine Viral Diarrhea Virus,BVDV)等其他病原;水样pH为8.73±0.14(轻微偏碱)、菌落总数达3.35×103CFU/mL(超标)并从中分离出短小芽孢杆菌(Bacillus pumilus,BP);饲料中呕吐毒素、玉米赤霉烯酮等均为阴性.现场调研发现腹泻犊牛与健康犊牛混饲、圈舍积水积粪、通风差、过度使用抗生素等环境和管理问题.确诊本次犊牛群发性腹泻以Cp感染为主,伴有饮水污染与环境管理不当所致.基于病因制定中西医结合治疗与综合预防方案,1个月后,犊牛腹泻发病率从40%降至10.58%.本研究明确了该场腹泻的病因,验证了减抗防治方案的有效性,为甘肃地区规模化奶牛场犊牛腹泻防控提供了实践参考.
To identify the etiological factors of calf diarrhea in a large-scale dairy farm in Lanzhou,Gansu Province,and provide a feasible anti-resistance prevention and control scheme,this study collected fecal samples from 19 diarrheic calves.Pathogen detection was conducted using colloidal gold technology,RT-qPCR,and saturated salt water flotation method.Drinking water samples were tested for pH,total bacterial colony count,and bacterial isolation and identification.Feed samples were analyzed for toxins via ELISA.The results showed that the nucleic acid positive rate of Clostridium perfringens(Cp)was 52.63%(10/19),and that of Cryptosporidium(COWP)was 5.26%(1/19).No other pathogens such as Bovine Rotavirus(BRV),Bovine Coronavirus(BCoV),or Bovine Viral Diarrhea Virus(BVDV)were detected.The drinking water was slightly alkaline(pH 8.73±0.14)with an excessive total bacterial colony count of 3.35×103 CFU/mL,from which Bacillus pumilus(BP)was isolated.Mycotoxins such as vomitoxin and zearalenone in feed were negative.On-site investigation revealed environmental and management issues including mixed feeding of diarrheic and healthy cattle,accumulated water and feces in pens,poor ventilation,and overuse of antibiotics.The outbreak was confirmed to be mainly caused by Cp infection,accompanied by drinking water contamination and improper environmental management.A comprehensive prevention and treatment scheme combining traditional Chinese and Western medicine was formulated.After one month,the total diarrhea incidence of the herd decreased from the initial 6.63%to 2.11%.This study clarified the etiological factors of diarrhea in the farm,verified the effectiveness of the anti-resistance and antibiotic-substituting prevention and control scheme,and provided practical references for diarrhea prevention and control in large-scale dairy farms in Gansu region.
