目的 分析分裂周期蛋白 20(CDC20)在肺腺癌(LUAD)组织中的表达及其与脑转移、临床病理特征的关系.方法 结合生物信息学分析与免疫组化的方法验证,通过UALCAN、GEPIA2、STRING及Kaplan-Meier Plotter平台,分析数据库中CDC20 mRNA的组织表达差异、TNM分期表达规律、与EMT通路蛋白互作关系及预后关联.以粤北人民医院 48 例LUAD患者的癌组织及对应癌旁组织为研究对象,采用免疫组化检测CDC20 蛋白表达,结合临床资料分析其与脑转移、TNM分期等指标的关系.结果 生信分析及免疫组化结果发现CDC20 mRNA及蛋白在LUAD组织中表达均高于癌旁组织(P<0.05);CDC20 高表达与LUAD患者TNM分期、脑转移相关(P<0.05),与年龄、性别、肿瘤位置无关(P>0.05),且与TNM分期、脑转移呈正相关(P<0.05);CDC20 与EMT通路核心分子存在直接蛋白互作关系(P<0.05),且CDC20 高表达患者的预后差于低表达患者.结论 CDC20 在LUAD组织中高表达,且临床分期及脑转移与CDC20 的高表达关系密切.

Objective To analyze the expression of cell division cycle protein 20(CDC20)in lung adenocarcinoma(LUAD)tissues and its relationships with brain metastasis and clinicopathological characteristics.Methods Combined with bioinformatics analysis and immunohistochemistry,the differences in tissue expression of CDC20 mRNA,TNM stage expression,interaction with EMT pathway proteins and prognosis were analyzed by UALCAN,GEPIA2,STRING and Kaplan-Meier Plotter platforms.The cancer tissues and corresponding adjacent tissues of 48 patients with LUAD in Yuebei People' s Hospital were taken as the research objects.The expression of CDC20 protein was detected by immunohistochemistry,and its relationship with brain metastasis and TNM staging was analyzed in combination with clinical data.Results The results of bioinformatics analysis and immunohistochemistry showed that the expression of CDC20 mRNA and protein in LUAD tissues was higher than that in adjacent tissues(P<0.05).The high expression of CDC20 was correlated with TNM stage and brain metastasis in LUAD patients(P<0.05),but not with age,gender and tumor location(P>0.05),and was positively correlated with TNM stage and brain metastasis(P<0.05).There was a direct protein interaction between CDC20 and the core molecules of EMT pathway(P<0.05),and the prognosis of patients with high expression of CDC20 was worse than that of patients with low expression.Conclusion CDC20 is highly expressed in LUAD tissues,and its high expression is closely associated with clinical stage and brain metastasis.
