超高效液相色谱-串联质谱法同时测定人全血中10种精神类药物浓度研究OA
Study on simultaneous determination of 10 antipsychotics in human whole blood by ultra-high performance liquid chromatography-tandem mass spectrometry
目的 建立一种快速、准确的超高效液相色谱-串联质谱(UHPLC-MS/MS)法检测人全血中甲基苯丙胺、氯胺酮、氯氮平等 10 种常见滥用药物及抗精神病药物浓度的分析方法.方法 血液样本采用乙腈蛋白沉淀法进行前处理;采用岛津 C18 色谱柱(2.1 mm×150 mm,3 μm)进行分离,以0.1%甲酸和20 mmol/L 乙酸铵水溶液以及乙腈体系进行等度洗脱,进样量 1 μL,流速 0.2 mL/min.采用内标定量法以电喷雾离子源(ESI),多反应监测(MRM)模式进行正离子扫描.总分析时长 4 min.同时对方法的特异性、线性范围、检出限、准确度与精密度、提取回收率、携带污染进行验证.结果 10 种药物在相应质量浓度范围内相关系数 r>0.9956,方法检出限为 2.0~3.5 ng/mL,定量下限为 10.00 ng/mL,在低、中、高不同添加浓度下准确度为 98.7%~110.6%,日内与日间精密度(RSD)均低于10.00%,提取回收率为 83.35%~110.95%.结论 本方法简便、快速、重现性好、检测限低、特异性强,适用于常见滥用药物及抗精神病药物浓度的快速定量分析.
Objective To establish a rapid and accurate ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)method for the determination of 10 common abused drugs and antipsychotics including meth-amphetamine,ketamine and clozapine in human whole blood.Methods Blood samples were pretreated using an acetonitrile protein precipitation method.Isocratic elution was carried out using the mobile phase system consisting of 0.1%formic acid containing 20 mmol/L ammonium acetate aqueous solution and acetonitrile.The injection volume was 1 μL with a flow rate of 0.2 mL/min.An electrospray ionization source(ESI)in positive ion mode and multiple reaction monitoring(MRM)scan-ning were applied,and internal standard quantification was used.The total analysis time was 4 min.Meanwhile,the specific-ity,linear range,limit of detection,accuracy and precision,extraction recovery and carryover contamination of the method were validated.Results The 10 drugs showed correlation coefficients(r)>0.995 within their respective concentration ran-ges.The method demonstrated limits of detection(LOD)of 2.0-3.5 ng/mL and lower limits of quantification(LLOQ)at 10.00 ng/mL.Accuracy ranged from 98.7%to 110.6%at low,medium,and high spiked concentrations,with both intra-day and inter-day precision(expressed as RSD)below 10.00%.Extraction recoveries were between 83.35%and 110.95%.Conclusion This method demonstrates simplicity,rapidity,good reproducibility,low detection limits,and strong specifici-ty.It is suitable for rapid quantitative analysis of common abused drugs and antipsychotics.
