清肝健脾活血方对CCl4诱导肝纤维化大鼠的保护作用研究OA
Prescription for clearing liver,fortifying spleen and resolving stasis alleviates liver injury in rats with CCl4-induced liver fibrosis
目的 探讨清肝健脾活血方对四氯化碳(CCl4)诱导肝纤维化大鼠的肝细胞保护作用.方法 将72 只雄性 SD 大鼠随机分成正常组、模型组、清肝健脾活血方高剂量组、清肝健脾活血方中剂量组、清肝健脾活血方低剂量组、鳖甲煎丸组,每组12 只.除正常组外,其余组大鼠采用腹腔注射40%CCl4 橄榄油混悬液方法诱导建立肝纤维化模型.于造模第2 天开始,清肝健脾活血方高、中、低剂量组分别按照32.084 g/kg、16.042 g/kg、8.021 g/kg 剂量给予清肝健脾活血方灌胃,鳖甲煎丸组按照0.9255 g/kg 剂量给予鳖甲煎丸灌胃,正常组和模型组给予纯净水灌胃,均1 次/d,连续灌胃8 周.HE 染色、Masson 染色、天狼星红染色观察肝组织病理形态,按照试剂盒方法测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、白蛋白(ALB)、乳酸脱氢酶(LDH)、总胆红素(TBil)、透明质酸(HA)、层黏连蛋白(LN)、Ⅲ型前胶原(PC-Ⅲ)、Ⅳ型胶原(Ⅳ-C)水平及肝组织中羟脯氨酸(Hyp)、谷胱甘肽过氧化物酶(GPX)、谷胱甘肽 S 转移酶(GST)含量,TUNEL 法检测肝细胞凋亡情况,Western blot 法检测肝组织中B 淋巴细胞瘤-2 相关X 蛋白(Bax)、半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)蛋白表达情况.结果 与正常组比较,模型组大鼠肝组织可见炎性细胞聚集,形成纤维间隔;血清 AST/ALT、LDH、TBil、HA、LN、PC-Ⅲ、Ⅳ-C 水平及肝组织中 Hyp、GST 含量均明显升高(P 均<0.05),血清 ALB 水平及肝组织中 GPX 含量均明显降低(P 均<0.05),细胞凋亡率和肝组织中 Bax、Caspase-3 蛋白相对表达量均明显升高(P 均<0.05).与模型组比较,清肝健脾活血方高剂量组和鳖甲煎丸组大鼠肝组织纤维化明显减轻,血清 AST/ALT、LDH、TBil、HA、LN、PC-Ⅲ、Ⅳ-C 水平及肝组织中 Hyp、GST 含量均明显降低(P 均<0.05),肝组织中 GPX 含量均明显升高(P 均<0.05),细胞凋亡率和肝组织中 Caspase-3 蛋白相对表达量均明显降低(P 均<0.05),且清肝健脾活血方高剂量组血清 ALB 水平明显升高(P<0.05),肝组织中 Bax 蛋白相对表达量明显降低(P<0.05).结论 清肝健脾活血方可明显减轻 CCl4 诱导的大鼠肝损伤,减少肝细胞凋亡,保护肝细胞.
Objective It is to observe the hepatoprotective effect of prescription for clearing liver,fortifying spleen and resolving stasis(PCLFSRS)on hepatocytes in rats with liver fibrosis induced by CCl4.Methods A total of 72 male SD rats were randomly divided into normal group,model group,high-dose,medium-dose and low-dose groups of PCLFSRS,Biejia-jian Pill(BJJP)group,with 6 rats in each group.The rats of all the groups except for the normal group were prepared with intraperitoneal injection of 40%CCl4 olive oil suspension to establish models of liver fibrosis.Starting from the 2rd day of modeling,high-dose,medium-dose and low-dose groups of PCLFSRS were administered PCLFSRS 32.084 g/kg,16.042 g/kg,and 8.021 g/kg via gavage,respectively,while the BJJP group was administered BJJP 0.9255 g/kg via gavage,the normal group and model group were given pure water via gavage,all once daily,continuously treated for 8 weeks.The pathological morphology of liver tissue was observed by HE staining,Masson staining,and Sirius red staining.The serum levels of ala-nine aminotransferase(ALT),aspartate aminotransferase(AST),albumin(ALB),lactate dehydrogenase(LDH),total bilirubin(TBil),hyaluronic acid(HA),laminin(LN),type Ⅲ procollagen(PC-Ⅲ),and type IV collagen(Ⅳ-C),as well as the contents of hydroxyproline(Hyp),glutathione peroxidase(GPX),and glutathione S-transferase(GST)in liver tissue were measured by the kit methods,the apoptosis of hepatocytes was assessed by TUNEL method,and the protein ex-pressions of Bax and Caspase-3 in liver tissue were detected by Western blot.Results Compared with the normal group,in-flammatory cell aggregation with fibrous septa formation in liver tissue were found in the rats of the model group,the serum levels of AST/ALT,LDH,TBil,HA,LN,PC-Ⅲ and Ⅳ-C,as well as then contents of Hyp and GST in liver tissue were significantly increased(all P<0.05),while the serum level of ALB and content of GPX in liver tissue were significantly decreased(all P<0.05),the apoptosis rate and relative protein expressions of Bax and Caspase-3 in liver tissue were sig-nificantly increased(all P<0.05).Compared with the model group,the fibrosis of liver tissue in the rats of the hight-dose group of PCLFSRS and BJJP group were obviously alleviated,the serum levels of AST/ALT,LDH,TBil,HA,LN,PC-Ⅲand Ⅳ-C,as well as then contents of Hyp and GST in liver tissue were significantly decreased(all P<0.05),while the contents of GPX in liver tissue were significantly decreased(all P<0.05),the apoptosis rates and relative protein expres-sions of Bax and Caspase-3 in liver tissue were significantly decreased(all P<0.05),the serum level of ALB was signifi-cantly increased,while the relative protein expression of Bax in liver tissue was significantly decreased in the high-dose group of PCLFSRS.Conclusion PCLFSRS can significantly alleviate CCl4 induced liver injury in rats,reduce liver cell ap-optosis,and protect liver cells.
