基于HPLC多成分定量与化学模式识别的不同产地防风质量差异分析OA
Analysis of quality variations in Radix Saposhnikoviae from different origins based on HPLC multi-component quantification combined with chemical pattern recognition
目的 明确不同产地防风色原酮类有效成分含量分布特征,揭示地理因素对其药材质量的影响.方法 采集黑龙江、内蒙古、河北、陕西、甘肃、河南 6 省区 18 批共 54 份防风样品,采用HPLC测定升麻素苷、升麻、5-O-甲基维斯阿米醇苷 3种色原酮类成分;并运用主成分分析(PCA)、层次聚类分析(HCA)、灰色关联度分析(GRA)开展多维度质量评价.结果 防风中 3 种色原酮类成分存在显著地理差异;其中,黑龙江、内蒙古产区防风样品里的升麻素苷、升麻、5-O-甲基维斯阿米醇三种色原酮成分含量,整体比河北、陕西、甘肃、河南产地的样品更高,其中升麻素苷含量区间为 2.09~6.12 mg/g,升麻为 1.37~6.12 mg/g,5-O-甲基维斯阿米醇苷为 1.40~3.74 mg/g.PCA 与 HCA 可将全部样品划分为 3 大类,东北产区单独归为高含量组别,野生、栽培防风成分含量差异具有统计学意义(P<0.05);灰色关联分析结果显示甘肃部分样品各类成分配比均衡,综合质量关联度更高.结论 防风色原酮类有效成分存在稳定地理分布规律,从化学成分层面佐证传统关防风道地优势,可为防风产地溯源、整体质量标准建立及优良栽培资源选育提供科学依据.
Objective To clarify the distribution characteristics of chromone constituents in Radix Saposhnikoviae from different ori-gins and to reveal the influence of geographic factors on its quality.Methods Fifty-four samples(18 batches)were collected from six provinces:Heilongjiang,Inner Mongolia,Hebei,Shaanxi,Gansu,and Henan.The contents of three chromones—prim-O-glucosylcimifugin,cimifugin,and 5-O-methylvisammioside—were determined by HPLC.Multidimensional quality evaluation was per-formed using principal component analysis(PCA),hierarchical cluster analysis(HCA),and grey relational analysis(GRA).Results Significant geographic variations were observed in the three chromone constituents.Samples from Heilongjiang and Inner Mongolia gen-erally showed higher contents of all three compounds than those from Hebei,Shaanxi,Gansu,and Henan.The content ranges were 2.09-6.12 mg/g for prim-O-glucosylcimifugin,1.37-6.12 mg/g for cimifugin,and 1.40-3.74 mg/g for 5-O-methylvisammioside.PCA and HCA classified all samples into three clusters,with the Northeastern origin forming a separate high-content group.Significant differences(P<0.05)were found between wild and cultivated samples.GRA indicated that some samples from Gansu had a more bal-anced proportion of constituents and thus higher comprehensive quality relational degrees.Conclusion The chromone constituents in Radix Saposhnikoviae exhibit stable geographic distribution patterns,providing chemical evidence for the superior quality of the tradi-tional"Guan Fangfeng(one variety from Northeast China)"and supporting origin traceability,quality standard establishment,and culti-var selection.
