目的 评价水洗工艺(掰瓣比、加水量、浸泡时间)对百合中王百合苷A与王百合苷B含量的影响,为百合地黄汤制备用百合的炮制工艺优化提供依据.方法 采用正交试验设计(因素水平:掰瓣比1/4~1瓣、加水量3~7倍、浸泡时间8~12 h),结合高效液相色谱法(HPLC)测定水洗前后百合中王百合苷A与王百合苷B的含量;色谱条件:ZORBAX SB-Aq色谱柱,乙腈-0.1%磷酸水(13:87)为流动相,检测波长312 nm,进一步采用方差分析评价水洗工艺参数对成分含量的影响.结果 未水洗百合(生百合)中王百合苷A与B含量分别为(3.075±0.060)mg·g⁻¹ 和(1.046±0.010)mg·g⁻¹;经不同参数水洗后,王百合苷A含量范围为2.938~3.068mg·g⁻¹,王百合苷B为1.027~1.044mg·g⁻¹,与未水洗组比较均无显著性差异(P>0.05);方差分析显示,掰瓣比、加水量、浸泡时间均对两种成分含量有显著影响(P<0.05或P<0.01),其中掰瓣比的影响最为显著;此外,王百合苷A与王百合苷B对不同水洗工艺参数的响应趋势存在一定的差异性.结论 在考察的水洗工艺参数范围内,尽管各参数对王百合苷A与王百合苷B含量有显著影响,但水洗处理本身并未导致二者显著损失,提示百合地黄汤制备用百合的传统水洗步骤对保留目标活性成分非必需.

Objective To evaluate the effects of water washing process parameters(bulb segmentation degree,water addition,and soaking time)on the contents of regaloside A and B in Bulbus Lilii,and to provide a basis for optimizing the processing technology of Bulbus Lilii for Baihe Dihuang Decoction(百合地黄汤,BHDHD).Methods An orthogonal experimental design(factor levels:bulb segmentation degree of 1/4 to 1 whole segment,water addition of 3-7 times,and soaking time of 8-12 h)was employed.High-performance liquid chromatography(HPLC)was used to determine the contents of regaloside A and B in Bulbus Lilii before and after water washing.The chromatographic separation was performed on a ZORBAX SB-Aq column with a mobile phase consisting of acetoni-trile-0.1%phosphoric acid aqueous solution(13:87,v/v)at a detection wavelength of 312 nm.The influence of the water washing pro-cess parameters on the contents of the two components was further evaluated by analysis of variance(ANOVA).Results The content of regaloside A and B in unwashed Bulbus Lilii was(3.075±0.060)mg·g-1 and(1.046±0.010)mg·g-1,respectively.After water washing with different parameters,the content of regaloside A ranged from 2.938 to 3.068 mg·g-1,and that of regaloside B ranged from 1.027 to 1.044 mg·g-1.No significant difference was observed compared with the unwashed group(P>0.05).The results of ANOVA showed that bulb segmentation degree,water addition,and soaking time all had significant effects on the contents of the two components(P<0.05 or P<0.01),with bulb segmentation degree being the most significant factor.Additionally,differences in the response trends of regaloside A and B to the different parameters were observed.Conclusion Within the range of water washing parameters investigated,although each factor had a significant effect on the contents of regaloside A and B,the water washing process itself did not result in sig-nificant losses of these compounds.This suggests that the traditional water washing step for Bulbus Lilii in the preparation of BHDHD is not necessary for retaining the target active ingredients.
