目的 研究茱萸丸对痤疮模型小鼠PPARγ/LXRα/ABCA1/CPT1α信号通路的影响,探讨其治疗痤疮的作用机制.方法 采用右耳耳廓涂抹油酸联合皮下注射痤疮丙酸杆菌诱导耳廓复合痤疮小鼠模型,造模周期为2周.造模成功的50只ICR小鼠随机分成5组,即模型组,茱萸丸低、中、高剂量组,异维A酸组各10只,另设10只同周龄健康ICR小鼠作为对照组,不造模.对照组与模型组予等体积无菌蒸馏水灌胃,茱萸丸低、中、高剂量组分别给予130.54、261.08、522.16 mg·kg-1灌胃,异维A酸组4.55 mg·kg-1灌胃,每日1次,各组小鼠共灌胃4周.干预后检测血清SOD、MDA、GSH-Px、CAT含量,进行耳廓组织HE、免疫荧光染色,Real-time PCR与Western blot检测PPARγ/LXRα/ABCA1/CPT1α相关基因和蛋白.结果 模型组小鼠耳廓组织病理改变加重;血清MDA含量升高,SOD、GSH-Px、CAT水平降低(P<0.01);耳廓组织PPARγ、LXRα、ABCA1、CPT1α蛋白及mRNA表达水平降低(P<0.01).与模型组比较,茱萸丸及异维A酸均可改善病理改变、氧化应激指标;耳廓组织PPARγ/LXRα/ABCA1/CPT1α通路表达水平升高(P<0.05,P<0.01).结论 茱萸丸可以改善痤疮模型小鼠炎性皮损,其机制可能与调控PPARγ/LXRα/ABCA1/CPT1α信号通路介导的氧化损伤及炎症反应有关.

Objective The study aimed to investigate the effects of Zhuyu Pill(朱黄丸,ZYP)on the PPARγ/LXRα/ABCA1/CPT1α signaling pathway in a mouse model of acne,and to explore its mechanism of action in treating acne.Methods A mouse model of acne was established by applying oleic acid on the right auricle and subcutaneous injection of Propionibacterium acnes to induce the auricle combined with acne.The modeling period was 2 weeks.Among the 50 successfully modeled ICR mice,they were randomly divided into 5 groups:the model group,the low,medium,and high doses of ZYP groups,and the isotretinoin group,with 10 mice in each group.Additionally,10 healthy ICR mice of the same age were set as the control group,without modeling.The control group and the model group were given equal volumes of sterile distilled water by gavage,while the low,medium,and high doses of ZYP groups were given 130.54,261.08,and 522.16 mg·kg-1·d-1 by gavage respectively,and the isotretinoin group was given 4.55 mg·kg-1·d-1 by gavage once a day.Each group of mice was gavaged for 4 weeks after the administration.After the intervention,the contents of serum SOD,MDA,GSH-Px and CAT were detected.HE staining,immunofluorescence staining of auricular tissue,real-time PCR and Western blot(WB)were used to detect the related genes and proteins of PPARγ/LXRα/ABCA1/CPT1α.Results The pathological changes of the auricle tissue in the model group mice were aggravated;the content of MDA in serum increased,while the levels of SOD,GSH-Px and CAT decreased(P<0.01);the expression levels of PPARγ,LXRα,ABCA1 and CPT1α protein and mRNA in auricle tissue decreased(P<0.01).Compared with the model group,both ZYP and Isotretinoin could improve the pathological changes and oxidative stress indi-cators;the expression levels of the PPARγ/LXRα/ABCA1/CPT1α pathway in auricle tissue increased(P<0.05,P<0.01).Conclusion ZYP can improve sebum secretion and inflammatory skin lesions in acne model mice,and its mechanism may be related to the activation of the PPARγ/LXRα/ABCA1/CPT1α signaling pathway.
