桃红四物汤对大鼠肢体缺血-再灌注损伤中miR-675、CaMKII、TNF-α的表达影响及干预作用OA
Effects of Taohong Siwu Decoction(桃红四物汤)on miR-675,CaMKII,and TNF-α expres-sion in rats with limb ischemia-reperfusion injury
目的 基于miR-675-CaMKII-炎症因子信号轴探讨桃红四物汤对肢体缺血-再灌注损伤的干预机制.方法 将雄性SD大鼠78只随机分为4组(对照组、模型组、miR-675阻断剂组和桃红四物汤组);除对照组外,余下各组均复制缺血-再灌注损伤模型;成功后选取各组0、2、4、8h时段取大鼠骨骼肌和血清做指标检测:HE染色法和DAPI法观察大鼠骨骼肌形态及病理性和细胞凋亡情况;RT-qPCR检测miR-675、CaMKIImRNA的表达,Western blot检测CaMKII蛋白表达,ELISA法检测血清TNF-α表达情况.结果 HE染色结果显示,模型组骨骼肌损伤最严重,miR-675阻断剂组和桃红四物汤组损伤较模型组减轻.DAPI荧光染色显示,模型组细胞凋亡数量最多,凋亡率高于其他各组,差异有统计学意义(P<0.05).RT-qPCR和Western blot的结果提示在同时段相比对照组:模型组、miR-675阻断剂组和桃红四物汤组的miR-675、CaMKII mRNA表达均增高,CaMKII蛋白的表达也增高(P<0.05);miR-675阻断剂组和桃红四物汤组的miR-675、CaMKII mRNA的表达均低于模型组,CaMKII蛋白的表达也降低(P<0.05),差异有统计学意义.ELISA血清检测提示各组TNF-α含量表达均高于对照组(P<0.05);miR-675阻断剂组和桃红四物汤组的TNF-α含量表达均低于模型组(P<0.05),差异有统计学意义.结论 桃红四物汤可以减轻大鼠骨骼肌细胞在肢体缺血-再灌注中的损伤,机制可能与桃红四物汤参与调控miR-675并进一步影响CaMKII蛋白、TNF-α等炎症因子的表达有关.
Objective To investigate the intervention effect of Taohong Siwu Decoction(桃红四物汤,THSWD)on the expressions of miR-675,CaMKII,and TNF-α in a rat model of limb ischemia-reperfusion injury(LIRI).Methods Seventy-eight male SD rats were randomly assigned to four groups:the control,model,miR-675 inhibitor,and THSWD groups.Except for the control group,LIRI was induced in the rats of the other groups.After successful modeling,skeletal muscle and serum samples were collected at 0,2,4,and 8 hours for analysis.HE staining was used to observe morphological and pathological changes in skeletal muscle,and DAPI staining was used to assess cell apoptosis.The expression levels of miR-675 and CaMKII mRNA were detected by RT-qPCR.CaMKII protein expres-sion was measured by Western blot(WB),and serum TNF-α level was measured by ELISA.Results HE staining revealed the most severe skeletal muscle injury in the model group,while the injury was alleviated in the miR-675 inhibitor and THSWD groups.DAPI staining indicated that the model group had the highest number of apoptotic cells and the highest apoptosis rate,which was significantly different from the other groups(P<0.05).At each corresponding time point,RT-qPCR and WB analyses showed that the expressions of miR-675 and CaMKII mRNA,as well as CaMKII protein,were significantly higher in the model,miR-675 inhibitor,and THSWD groups than in the control group(P<0.05).However,the expressions in the miR-675 inhibitor and THSWD groups were significantly lower than those in the model group(P<0.05).ELISA revealed that serum TNF-α levels in all experimental groups were higher than those in the control group(P<0.05).Notably,the levels in the miR-675 inhibitor and THSWD groups were significantly lower than those in the model group(P<0.05).Conclusion THSWD alleviates LIRI in rat skeletal muscle.The mechanism may be associated with its regulation of miR-675 expression,thereby modulating the expression of CaMKII and TNF-α and other inflammatory factors.
