基于Wnt/β-catenin信号通路探讨搜风愈喘方改善哮喘大鼠气道重塑的作用机制OA
Exploring the mechanism of Soufeng Yuchuan Formula(搜风愈喘方)in ameliorating airway remodeling in asthmatic rats via the Wnt/β-catenin signaling pathway
目的 探讨搜风愈喘方对哮喘模型大鼠气道重塑的影响,阐明其通过Wnt/β-catenin信号通路介导上皮间质转化的作用机制.方法 将48只SPF级6~8 周龄大鼠随机分为空白组、模型组、地塞米松组及搜风愈喘方低、中、高剂量组,每组8只.卵清蛋白和氢氧化铝凝胶建立哮喘模型后,给药3周取材.观察大鼠一般情况,HE及Masson染色观察肺组织病理学变化;ELISA检测血清IFN-γ、IL-4、IL-1β、MMP-9含量;RT-PCR检测肺组织Wnt5a、Wnt7a、β-catenin、GSK-3β、C-Myc、E-cadherin mRNA表达;Western blot检测肺组织Wnt5a、β-catenin、GSK-3β、Vimentin蛋白表达.结果 与模型组比较,搜风愈喘方组大鼠体质量增加(P<0.05,P<0.01),抓鼻、喘息等症状改善(P<0.01);气道炎症评分及胶原沉积率降低(P<0.01);血清 IFN-γ升高(P<0.05,P<0.01),IL-4、IL-1β、MMP-9降低(P<0.01);肺组织 Wnt5a、Wnt7a、β-catenin、GSK-3β、C-Myc mRNA 表达降低(P<0.05,P<0.01),E-cadherin mRNA升高(P<0.05,P<0.01);Wnt5a、β-catenin、GSK-3β、Vimentin蛋白表达降低(P<0.01).结论 搜风愈喘方可能通过调控Wnt/β-catenin信号通路影响上皮间质转化,改善气道重塑以治疗哮喘.
Objective To explore the effect of the Soufeng Yuchuan Formula(搜风愈喘方,SFYCF)on airway remodeling in asthma model rats and clarify its mechanism of mediating epithelial-mesenchymal transition(EMT)by regulating the Wnt/β-catenin signaling pathway.Methods Forty-eight 6-8-week-old specific pathogen free(SPF)rats were randomly divided into the blank group,the model group,the dexamethasone group,the low/medium/high-dose SFYCF groups,with 8 rats in each group.After establishing an asthma model with aluminum hydroxide adjuvant,the rats were administered with corresponding drugs for 3 weeks before sampling.The general conditions of the rats were observed,and HE and Masson staining were used to observe lung tissue pathology.The levels of IFN-γ,IL-4,IL-1β,and MMP-9 in sera were detected by ELISA.RT-PCR was used to detect the mRNA expressions of Wnt5a,Wnt7a,β-catenin,GSK-3β,C-Myc,and E-cadherin in the lung tissue,while Western blot(WB)was used to detect the protein expressions of Wnt5a,β-catenin,GSK-3β,and Vimentin in the lung tissue.Results Compared with the model group,the body weight of rats in the SFYCF groups was increased(P<0.05,P<0.01),and their symptom scores such as nose scratching and wheezing were improved(P<0.01);airway inflammation scores and collagen deposition rates were decreased(P<0.01);serum IFN-γ levels were increased(P<0.05,P<0.01),while IL-4,IL-1β,and MMP-9 levels were decreased(P<0.01);mRNA expressions of Wnt5a,Wnt7a,β-catenin,GSK-3β,and C-Myc in lung tissue were decreased(P<0.05,P<0.01),while E-cadherin mRNA expression was increased(P<0.05,P<0.01);protein expressions of Wnt5a,β-catenin,GSK-3β,and Vimentin were decreased(P<0.01).Conclusion SFYCF may affect EMT by regulating the Wnt/β-catenin signaling pathway,thereby ameliorating airway remodeling and exerting a therapeutic effect on asthma.
