通络蠲痹颗粒调控ROS-NLRP3-Caspase-1轴对骨关节炎软骨细胞焦亡的影响OA
Effects of Tongluo Juanbi Granule(通络蠲痹颗粒)on pyroptosis of osteoarthritis chondro-cytes via the ROS-NLRP3-Caspase1 axis
目的 从细胞水平探究通络蠲痹颗粒(TLJB)含药血清对兔软骨细胞焦亡及活性氧自由基(ROS)-NOD 样受体蛋白3(NLRP3)-胱天蛋白酶-1(Caspase-1)信号轴的影响,研究其防治骨关节炎(OA)的作用机制.方法 提取TLJB含药血清及空白血清,体外培养兔软骨细胞,分为正常对照组、模型组、空白血清组、中药含药血清组、NLRP3激活剂组、中药+NLRP3激活剂组.甲苯胺蓝染色法(Toluidine Blue)及Ⅱ型胶原免疫荧光法鉴定软骨细胞;噻唑蓝法(MTT比色法)筛选最佳中药含药血清干预浓度;偶联酶反应法检测乳酸脱氢酶(LDH)水平;酶联免疫吸附测定(ELISA)检测IL-1β、IL-18水平;化学发光法检测ROS水平;实时荧光定量PCR(RT-qPCR)检测软骨细胞NLRP3、Caspase-1、GSDMD、IL-1β、IL-18的mRNA表达,蛋白免疫印迹(WB)检测NLRP3、pro-Caspase-1、GSDMD的蛋白表达.结果 经MTT法检测15%TLJB含药血清浓度干预48h时最适合兔软骨细胞增殖.与正常对照组比较,模型组LDH、IL-1β、IL-18、ROS及NLRP3、Caspase-1、GSDMD mRNA和蛋白表达均显著升高(P<0.01);与模型组比较,NLRP3激活剂组LDH、IL-1β、IL-18、ROS及NLRP3、Caspase-1、GSDMD mRNA和蛋白表达均显著升高(P<0.01),中药含药血清组LDH、IL-1β、IL-18、ROS及NLRP3、Caspase-1、GSDMD mRNA和蛋白表达均显著降低(P<0.01);与NLRP3激活剂组比较,中药+NLRP3激活剂组LDH、IL-1β、IL-18、ROS及NLRP3、Caspase-1、GSDMD mRNA和蛋白表达均显著降低(P<0.01).结论 TLJB含药血清可抑制兔关节软骨细胞焦亡,其作用机制可能与抑制ROS-NLRP3-Caspase1信号轴的激活有关.
Objective To explore the effects of Tongluo Juanbi Granules(通络蠲痹颗粒,TLJB)medicated serum on pyroptosis and the reactive oxygen species(ROS)-NOD-like receptor protein 3(NLRP3)-Caspase-1 signaling axis in rabbit chondrocytes at the cellu-lar level,and to study its mechanism of action in preventing and treating osteoarthritis(OA).Methods TLJB medicated serum and blank serum were extracted.Rabbit chondrocytes were cultured in vitro,and divided into normal control group(group A),model group(group B),blank serum group(group C),TLJB medicated serum group(group D),NLRP3 activator group(group E),and TLJB medi-cated serum+NLRP3 activator group(group F).Chondrocytes were identified using Toluidine Blue staining and Type II collagen immu-nofluorescence.MTT assay was used to select the optimal intervention concentration of TLJB medicated serum.Lactate dehydrogenase(LDH)levels were detected by a coupled enzyme assay.Levels of IL-1β and IL-18 were detected by enzyme-linked immunosorbent assay(ELISA).ROS levels were detected by a chemiluminescence method.Real-time quantitative PCR(RT-qPCR)was used to detect the mRNA expression of NLRP3,Caspase-1,GSDMD,IL-1β and IL-18 in chondrocytes.Western blot(WB)was used to detect the protein expression of NLRP3,pro-Caspase-1 and GSDMD.Results MTT assay showed that 15%TLJB medicated serum intervention for 48 hours was optimal for rabbit chondrocyte proliferation.Compared with group A,LDH,IL-1β,IL-18,ROS levels,and mRNA and protein expression of NLRP3,Caspase-1,and GSDMD in group B were significantly increased(P<0.01).Compared with group B,group E showed significant increases in LDH,IL-1β,IL-18,ROS levels,and mRNA and protein expression of NLRP3,Caspase-1,and GSDMD(P<0.01),while group D showed significant decreases in these indicators(P<0.01).Compared with group E,LDH,IL-1β,IL-18,ROS levels,and mRNA and protein expression of NLRP3,Caspase-1,and GSDMD in group F were significantly decreased(P<0.01).Conclusion TLJB medicated serum can inhibit pyroptosis in rabbit articular chondrocytes,and its mechanism may be related to the suppression of the ROS-NLRP3-Caspase1 signaling axis activation.
