circR-6041对阴道润滑障碍的影响及其机制OA
Effect of circR-6041 on vaginal lubrication disorder and its underlying mechanism
目的 基于MAPK/ERK信号通路探索circR-6041在阴道上皮功能障碍中的作用机制.方法 建立circR-6041过表达的VK2/E6E7细胞模型,采用划痕实验、Transwell实验和CCK-8检测细胞迁移和增殖能力.转录组测序分析circR-6041过表达后差异表达基因(DEGs),KEGG富集分析DEGs的富集通路,采用Western blot检测MAPK/ERK信号通路关键蛋白的表达水平.结果 过表达circR-6041显著增强VK2/E6E7细胞迁移能力(P<0.05),但不影响增殖(P>0.05).转录组和KEGG富集分析显示,circR-6041过表达组与对照组的差异基因显著富集于MAPK、PI3K-Akt等信号通路以及细胞外基质、紧密连接等细胞组分.Western blot结果显示,过表达circR-6041后,p-MEK1和p-ERK1/2蛋白表达增加(P<0.05),而总蛋白水平无明显变化,p-MEK1/MEK1和p-ERK1/2/ERK1/2比值升高(P<0.05).结论 circR-6041在阴道润滑障碍(LD)患者阴道上皮中表达上调,可能通过激活MAPK/ERK信号通路促进细胞迁移,参与LD的病理过程.
Objective To investigate the role and mechanism of circR-6041 in vaginal epithelial dysfunction based on the MAPK/ERK signaling pathway.Methods A VK2/E6E7 cell model overexpressing circR-6041 was established.Cell migration,invasion,and proliferation capabilities were assessed using wound healing assay,Transwell assay,and CCK-8 assay,respectively.Differen-tially expressed genes(DEGs)were screened after circR-6041 overexpression by transcriptomic sequencing(RNA-seq),and DEGs were analyzed by KEGG.Western blot was employed to detect the expressions of key proteins in the MAPK/ERK signaling pathway.Results Overexpression of circR-6041 significantly enhanced the migration ability of VK2/E6E7 cells(P<0.05),but did not affect the proliferation(P>0.05).Transcriptomic and KEGG enrichment analyses revealed that DEGs between circR-6041 overexpression group and control group were significantly enriched in signaling pathways such as MAPK and PI3K-Akt,and cellular components such as extracellular matrix(ECM)and tight junctions.Western blot results demonstrated that circR-6041 overexpression up-regulated the protein expressions of p-MEK1 and p-ERK1/2(P<0.05),total protein levels remained unchanged,and the ratios of p-MEK1/MEK1 and p-ERK1/2/ERK1/2 significantly increased(P<0.05).Conclusion circR-6041 is up-regulated in the vaginal epithelium of patients with lubrication disorders(LD),and circR-6041 can participate in the pathological process of LD by promoting cell migration via the activation of MAPK/ERK signaling pathway.
