目的 基于蛋白质组学技术比较不同诱导条件下中性粒细胞胞外捕获网(NETs)的异同,阐明其共性核心机制与特异性调控通路,为精准靶向肿瘤相关NETs提供实验依据.方法 分离BALB/c小鼠骨髓中性粒细胞,分别以佛波酯(PMA)、肿瘤培养上清(TCM)诱导NETs形成,采用免疫荧光染色验证NETs生成;通过非靶向定量蛋白质组学检测蛋白表达谱;运用生物信息学分析差异蛋白/共同蛋白的亚细胞定位、GO注释、KEGG通路富集及蛋白互作网络分析.结果 PMA和TCM均可成功诱导NETs;两组共鉴定出8 966种共同蛋白,富含组蛋白、中性粒细胞弹性蛋白酶(NE)、髓过氧化物酶(MPO)等经典NETs标志物,定位于细胞质与细胞膜,主导基础代谢、物质转运与结构组装.两组差异蛋白共2 765种,PMA-NETs高表达蛋白聚焦氧化磷酸化、活性氧(ROS)生成、吞噬体通路;而TCM-NETs高表达蛋白富集细胞周期、DNA修复、代谢重塑、补体凝血及肿瘤相关炎症通路.结论 PMA与TCM诱导的NETs共享保守核心蛋白与基础通路,同时又有明显的诱导条件特异性,二者同中有异;TCM诱导的NETs呈现出更贴近肿瘤微环境的表型倾向.这为区分经典化学诱导型NETs与肿瘤相关NETs的分子特征、开发针对性的靶向策略提供了理论依据.

Objective To compare the differences and similarities of neutrophil extracellular traps(NETs)under various induction conditions based on proteomic techniques,and elucidate their core common mechanisms and specific regulatory pathways for providing experimental evidence in the precise targeting of tumor-associated NETs.Methods Bone marrow neutrophils were isolated from BALB/c mice and induced to form NETs using phorbol 12-myristate 13-acetate(PMA)or tumor cell-conditioned medium(TCM).NETs were verified by immunofluorescence staining.Protein expression profiles were detected using untargeted quantitative proteomics.Bioinformatics analyses were performed to assess the subcellular localization,Gene Ontology(GO)functional enrichment,Kyoto Ency-clopedia of Genes and Genomes(KEGG)pathway enrichment and protein-protein interaction network analysis of common proteins and differential proteins.Results Both PMA and TCM successfully induced NETs.A total of 8 966 common proteins were identified in both two groups,including classical NET markers such as histone,neutrophil elastase(NE),and myeloperoxidase(MPO).These proteins were primarily localized in the cytoplasm and cell membrane,and involved in basic metabolism,substance transport,and structural assembly.A total of 2 765 differential proteins were identified between the two groups.Proteins up-regulated in PMA-induced NETs were enriched in oxidative phosphorylation,reactive oxygen species production,and phagosome pathways.Proteins up-regulated in TCM-induced NETs were enriched in cell cycle regulation,DNA repair,metabolic reprogramming,complement-coagulation cascades,and tumor-associated inflammatory pathways.Conclusion PMA-and TCM-induced NETs share conserved core proteins and basic pathways while exhibiting significant stimulus-specific features,presenting a"conserved yet distinct"profile.TCM-induced NETs tend to develop an adaptive phenotype tailored to the tumor microenvironment.This study provides a theoretical basis for distinguishing the molecular characteristics of classical chemical-induced NETs from tumor-associated NETs and developing targeted therapeutic strategies.
