蠲痹汤加减对胶原诱导性关节炎大鼠作用机制OA
Mechanism of action of modified Juanbi decoction on collagen-induced arthritis rats
目的:观察蠲痹汤加减对Ⅱ型胶原诱导性关节炎(CIA)大鼠的作用机制.方法:选取SPF级SD大鼠30只,随机分为对照组、模型组、雷公藤多苷组、蠲痹汤加减高剂量组、蠲痹汤加减低剂量组,每组6只.采用牛Ⅱ型胶原与弗氏完全佐剂乳化液诱导建立CIA模型.造模成功后,灌胃给药每日1次,连续4周.动态测量足踝肿胀度;末次给药后,检测全血黏度、血沉等血液流变学指标;称量脾脏及胸腺湿重,计算脏器系数;流式细胞术检测外周血T淋巴细胞亚群(CD3+、CD4+、CD8+);ELISA检测血清肿瘤坏死因子-α(TNF-α)及白细胞介素-1β(IL-1β)水平;HE染色观察踝关节滑膜及软骨组织病理学变化.结果:与模型组相比,蠲痹汤加减能显著降低CIA大鼠足踝关节肿胀的宽度与厚度(P<0.05),改善关节滑膜增生、炎性细胞浸润等病理损伤;降低高、中、低切变率下的全血黏度、红细胞聚集指数及血沉(P<0.05);抑制脾脏及胸腺肿大(P<0.05);回调CD4+/CD8+比值,下调血清TNF-α、IL-1β水平(P<0.05).结论:本研究以"活血-免疫-抗炎"三维调控网络,揭示蠲痹汤加减"扶正解毒、通络除痹"的作用,诠释蠲痹汤加减治疗类风湿关节炎机制.
Objective:To investigate mechanism of action of modified Juanbi decoction on type Ⅱ collagen-in-duced arthritis(CIA)in rats.Methods:Thirty SPF-grade SD rats were selected and randomly divided into blank con-trol group,model group,tripterygium glycosides group,and two dosage groups of modified Juanbi decoction(high,medium,and low doses).Each group consisted of 6 rats.The CIA model was established by inducing with bovine type Ⅱ collagen and Ficoll complete adjuvant emulsion.After successful modeling,the rats were administered intra-gastrically once daily for 4 weeks.The swelling degree of the ankle joint was measured dynamically;after the last ad-ministration,the blood rheological parameters such as whole blood viscosity,erythrocyte aggregation index,and erythrocyte sedimentation rate were detected;the wet weights of the spleen and thymus were weighed,and the organ coefficients were calculated;the peripheral blood T lymphocyte subsets(CD3+,CD4+,CD8+)were detected by flow cytometry;the serum levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were detected by ELISA;the pathological changes of the ankle joint synovium and cartilage tissue were observed by HE staining.Results:Com-pared with the model group,modified Juanbi decoction significantly reduced the width and thickness of the ankle joint swelling in CIA rats(P<0.05),improved the pathological damage such as synovial hyperplasia and inflammatory cell infiltration;significantly reduced the whole blood viscosity,erythrocyte aggregation index,and erythrocyte sedi-mentation rate at high,medium,and low shear rates(P<0.05);inhibited the enlargement of the spleen and thymus(P<0.05);significantly adjusted the CD4+/CD8+ratio and downregulated the serum levels of TNF-α and IL-1β(P<0.05).Conclusion:This study,based on the"blood-activation-immune-inflammation"three-dimensional regula-tory network,reveals the"strengthening the body,detoxifying,and promoting circulation to relieve arthralgia"effect of modified Juanbi decoction,and explaining mechanism of modified Juanbi decoction in the treatment of rheumatoid arthritis.
