目的 比较鲜铁皮石斛经鲜榨、鲜榨回流两种工艺制备的提取物与鲜铁皮石斛烘干后经回流制备的提取物(即干品回流)的药效差异,筛选鲜铁皮石斛最佳制备工艺,促进中药鲜药制剂的研发和推广应用.方法 分别以鲜榨、鲜榨回流、干品回流3 种工艺制备铁皮石斛提取物,分为高(8g·kg-1)、中(4g·kg-1)、低(2g·kg-1)剂量,通过乙醇性胃溃疡、燥结便秘、阿托品致胃肠运动障碍及醋酸扭体疼痛 4 种小鼠模型,对比铁皮石斛不同工艺提取物的药效差异.结果 3 种提取物均能减轻乙醇性胃溃疡,药物组溃疡指数均低于模型组(P<0.05 或 P<0.01),其中鲜榨高剂量组的溃疡指数低于干品高剂量组(P<0.05);燥结便秘排便实验中,3 种提取物的促排便作用都存在一定剂量相关趋势,其中鲜榨高剂量组的粪便粒数、粪便质量高于模型组(P<0.05);胃肠运动障碍模型中,仅鲜榨可促进小鼠胃排空(P<0.05);3 种铁皮石斛提取物均无明显镇痛作用.结论 鲜铁皮石斛的总体药效优于干品,其中鲜榨工艺的抗胃溃疡、促胃肠运动作用最优,为鲜铁皮石斛制剂的进一步推广应用,提供了可使其发挥最佳药效的提取工艺.

Objective To compare the pharmacological effects of extracts of fresh Dendrobium officinale Kimura&Migo(D.officinale)via two processes(freshly squeezed,freshly squeezed reflux)and the extract from dried D.officinale(prepared by reflux process),screen the optimal preparation technology,and promote the popularization and application of fresh herb preparation.Methods Three distinct extracts of D.officinale were prepared:freshly squeezed extract,freshly squeezed reflux extract,and dried product reflux extract,with three dosage levels:high(8 g·kg-1),medium(4 g·kg-1),and low(2 g·kg-1).The differences in pharmacological effects among these extracts were evaluated using four animal models in mice:ethanol-induced gastric ulcer,constipation,atropine-induced gastrointestinal motility disorder,and acetic acid-induced writhing pain.Results All three extracts alleviated ethanol-induced gastric ulcers,with the ulcer index of drug-treated groups significantly lower than that of model group(P<0.05 or P<0.01).The ulcer index in high-dose freshly squeezed group was significantly better than that in high-dose dried product group(P<0.05).In the defecation experiment with constipated mice(due to dryness and stagnation),the laxative effects of the three extracts showed a certain dose related trend,the number and weight of feces in high-dose freshly squeezed group were significantly higher than those in model group(P<0.05);In the gastrointestinal motility disorder model,only the freshly squeezed concentration extract could promote gastric emptying in mice(P<0.05).Notably,none of the three extracts of D.officinale showed significant analgesic effect.Conclusion The pharmacological effects of fresh D.officinale are superior to those of the dried product.Among the tested extraction processes,the freshly squeezed method yields the optimal effects in anti-gastric ulcer and promoting gastrointestinal motility.This finding provides the optimal extraction process for exerting the pharmacological effects of fresh D.officinale,thereby facilitating its further popularization and application.
