含自然铜中药复方对SD大鼠的毒性病理学研究OA
Toxicopathological study of pyrite-containing traditional Chinese medicine compound on SD rats
目的 研究含自然铜中药复方 YKY-018 长期使用的安全性及其所含铜在重要脏器的毒性病理学变化.方法 将 SD 大鼠随机分为溶媒对照组和含自然铜中药复方 YKY-018 低、中、高剂量组(0.25、0.5、1g·kg-1 剂量组),经口灌胃,每天给药 1 次,连续给药 13 周,停药恢复 4 周.通过肉眼形态学、脏器重量、脏脑比、苏木精-伊红(HE)染色及铜元素特殊染色(红氨酸法)的方法进行病理学评价.结果 肉眼形态学观察,高剂量组肾脏体积大,肾脏脏器重量和脏脑比升高.高剂量组雄性大鼠肝脏脏器重量和脏脑比升高.HE 检查显示,低剂量组肾脏轻微肾小管肥大,中、高剂量组肾脏轻微至轻度肾小管肥大,其他组织脏器均未见异常.红氨酸法检测,高剂量组肾脏肾小管上皮细胞内可见铜元素.停药 4 周后,肾脏病变恢复正常.结论 YKY-018 连续灌胃给予大鼠 13 周,0.25、0.5、1g·kg-1剂量可引起剂量相关性的轻微至轻度肾小管上皮细胞肥大,以上病理学改变停药 4 周后可恢复正常.
Objective To investigate the long-term safety of YKY-018,a traditional Chinese medicine(TCM)compound containing pyritum,as well as the toxicopathological changes of copper in vital organs induced by this compound.Methods Sprague-Dawley(SD)rats were randomly divided into the vehicle control group and the low-,medium-,and high-dose YKY-018 groups(0.25,0.5 and 1 g·kg-1,respectively).The rats were administered the drug via oral gavage once daily for 13 consecutive weeks,followed by a 4-week recovery period after drug with drawal.Pathological evaluations were conducted by means of macroscopic morphological observation,organ weight measurement,organ-brain ratio calculation,hematoxylin-eosin(HE)staining,and specific staining for copper(rubeanic acid method).Results Macroscopic morphological observation showed that the high-dose group presented an enlarged kidney volume,along with significant increases in kidney weight and kidney-brain ratio.In male rats of the high-dose group,the liver weight and liver-brain ratio also increased significantly.HE staining revealed slight renal tubular hypertrophy in the low-dose group,and slight to mild renal tubular hypertrophy in the medium-and high-dose groups,while no abnormalities were detected in other tissues and organs.Rubeanic acid staining demonstrated the presence of copper in the renal tubular epithelial cells of the high-dose group.After the 4-week recovery period,all the aforementioned renal lesions returned to normal.Conclusion Following 13 weeks of repeated oral gavage administration of YKY-018 to rats at doses of 0.25,0.5,and 1 g·kg-1,dose-related minimal to slight hypertrophy of the renal tubular epithelial cells was observed.These pathological changes were reversible to normal levels after a 4-week recovery period.
