目的 确定复方甲氨蝶呤-氨茶碱水凝胶的制备工艺处方,并探究其对寻常型银屑病小鼠模型的治疗效果.方法 采用单因素与效应面(Box-Behnken)法优化制剂处方及工艺条件.将 15 只小鼠随机分为 3 组,其中一组不做处理为对照组,其他两组连续 6d 在左耳涂抹 5%咪喹莫特(IMQ)乳膏建立银屑病样皮损模型,并分为对照组、银屑病模型组、复方甲氨蝶呤-氨茶碱水凝胶组.观察小鼠耳部皮损,记录耳部厚度变化,对耳部皮肤进行 HE染色观察病理变化.采用 Western blotting 检测耳部皮肤组织 JAK1/STAT3/STAT5 蛋白表达水平.结果 复方甲氨蝶呤-氨茶碱水凝胶最优处方工艺条件为:卡波姆 940 用量 2.04%,丙二醇用量 21.49%,氮酮用量 2.85%,体系pH 为 7.0.复方甲氨蝶 呤-氨茶碱水凝胶组小鼠皮损减轻,左耳厚度减小,皮肤表皮厚度减小,耳部皮肤组织JAK1、STAT3、STAT5 的蛋白相对表达量降低.结论 确定的水凝胶制备工艺相对简单,重复性良好.复方甲氨蝶呤-氨茶碱水凝胶对银屑病小鼠模型治疗效果良好,减轻皮肤炎症,可能通过 JAK/STAT 炎症信号通路发挥治疗作用.

Objective To optimize the formulation and preparation technology of compound methotrexate-aminophylline hydrogel,and explore its therapeutic effect on mice model of vulgaris psoriasis.Methods Single factor test and Box-Behnken response surface methodology were applied to optimize the formulation and preparation parameters.A total of 15 mice were randomly divided into three groups,among which one group was set as blank control group without any treatment.The other two groups were topically administered with 5%imiquimod(IMQ)cream on the left ear for 6 consecutive days to establish psoriasis-like skin lesion models,namely the psoriasis model group and the compound methotrexate-aminophylline hydrogel treatment group.The ear skin lesions of mice were observed,and the changes in ear thickness were recorded.HE staining was performed to observe the pathological changes of ear skin and small intestinal tissues.Western blotting was used to detect the protein expression levels of JAK1,STAT3 and STAT5 in ear skin tissues.Results The optimal formulation and preparation conditions of compound methotrexate-aminophylline hydrogel were as follows:Carbomer 940 2.04%,propylene glycol 21.49%,azone 2.85%,and the system pH was 7.0.In the compound methotrexate-aminophylline hydrogel group,the skin lesions of mice were significantly alleviated,the thickness of left ear and epidermal thickness were reduced,and the relative protein expression levels of JAK1,STAT3 and STAT5 in ear skin tissues were down-regulated.Conclusion The established hydrogel preparation process is relatively simple and demonstrates reproducibility.Compound methotrexate-aminophylline hydrogel has a good therapeutic effect on psoriasis mouse model,reducing skin inflammation,which may play a therapeutic role through JAK/STAT inflammatory signal pathway.
