重唇石斛根的化学成分及其生物活性研究OA

为探讨重唇石斛(Dendrobium hercoglossum)根部位的化学成分及其生物活性,对其根部位乙醇提取物的乙酸乙酯相用硅胶柱层析、半制备液相色谱等现代色谱技术进行分离纯化,再根据质谱和核磁等技术对单体化合物进行结构鉴定和抗炎活性筛选.结果表明,从重唇石斛根部位乙醇提取物乙酸乙酯相共分离鉴定了8个单体化合物,分别为(Z)-烯酰吗啉(1)、(E)-烯酰吗啉(2)、dengibsin(3)、4-O-demethyl-nobilone(4)、dendroflorin(5)、对羟基苯丙酸(6)、丁香酸(7)、1H-indole-3-carbal-dehyde(8).活性测试结果表明化合物4能够抑制LPS诱导小鼠单核巨噬细胞RAW264.7产生NO,IC50值为(14.69±1.16)μmol/L.化合物1 和2 为首次从自然界中分离得到,并首次报道其核磁数据;化合物1~8 均为首次从重唇石斛中分离得到,且化合物4 具有较好的抗炎活性.

To investigate the chemical constituents of the roots of Dendrobium hercoglossum and evaluate their biological activities,the ethyl acetate fraction of an ethanol extract from its roots was isolated and purified by using chromatographic techniques,including silica gel column chromatography and semi-preparative HPLC.The structures of the isolated compounds were elucidated through mass spectrometry and nuclear magnetic resonance spectroscopy(NMR),and then they were subsequently evaluated for anti-inflammatory and other biological activities.The results showed that eight compounds were isolated,such as(Z)-dimethomorph(1),(E)-dimethomorph(2),dengibsin(3),4-O-demethyl-nobilone(4),dendroflorin(5),p-hydroxyphenylpropionic acid(6),syringic acid(7),1H-indole-3-carbaldehyde(8).Compound 4 significantly inhibited LPS-induced NO production in RAW264.7 mouse macrophages,with an IC₅₀ value of(14.69±1.16)μmol/L.Compounds 1 and 2 were isolated as natural products for the first time,as well as their NMR data.Compounds 1-8 were isolated from D.hercoglossum for the first time.Compound 4 showed significant anti-inflammatory activity.
