丁香与母丁香中丁香酚及母丁香酚的提取工艺优化OA
Process Optimization for Extraction of Eugenol and Bancroftinone from Eugenia caryophyllata Thunb.and Syzygium aromaticum(L.)Merr.&L.M.Perry
[目的]筛选获得丁香、母丁香提取物有效成分含量高、适用于工业化大规模生产、操作简便且成本可控的提取工艺,为其后续制剂研发及药理研究提供技术支撑.[方法]以提取物中丁香酚及母丁香酚的含量(即提取物得酚率)为核心评价指标,采用高效液相色谱法(HPLC)系统比较水蒸气蒸馏法、超声提取法、索氏提取法、超声联合酶辅助蒸馏法4种提取方法的工艺效率,筛选出最佳提取方法.在此基础上,通过粉碎粒度、料液比、浸泡时间三因素的单因素试验结合L9(34)正交试验设计,对水蒸气蒸馏工艺进行优化,并对最优提取工艺进行验证.[结果]对比4种提取方法,水蒸气蒸馏法所得提取物的得酚率显著高于其他提取方法.正交试验方差分析结果表明,粉碎粒度、料液比、浸泡时间3个因素在试验设定梯度内对得酚率无显著影响(P>0.05,α=0.05);结合正交试验结果及工业化生产能耗综合考量,确立丁香水蒸气蒸馏最佳工艺条件:粉碎粒度80目、料液比1∶14(g/mL)、浸泡时间6 h;母丁香最佳工艺条件为:粉碎粒度80目、料液比1∶12(g/mL)、浸泡时间24 h.[结论]验证试验证实,在上述优化的最佳工艺条件下,丁香、母丁香提取物中目标有效成分含量最高.该工艺稳定、重复性好,可作为丁香及母丁香高效提取的标准化工业工艺.
[Objective]To obtain an extraction process with high content of effective components,suitable for large-scale industrial production,simple operation,and controllable cost,so as to provide technical support for subsequent formulation development and pharmacological research of Eugenia caryophyllata Thunb.and Syzygium aromaticum(L.)Merr.&L.M.Perry extracts.[Method]Taking the content of eugenol and bancroftinone in the extract(i.e.,the extract phenol rate)as the core evaluation index,the process efficiency of four extraction methods:water vapor distillation,ultrasonic extraction,Soxhlet extraction,and ultrasonic-assisted distillation with enzyme assistance,was compared using high-performance liquid chromatography(HPLC).The best extraction method was selected.Based on this,by single-factor experiments of particle size,liquid-to-solid ratio,and soaking time combined with L9(34)orthogonal experimental design,the water vapor distillation process was optimized,and the optimal extraction process was verified.[Result]By comparison of the four extraction methods,the extract obtained by water vapor distillation had a significantly higher phenol content than those of the other traditional methods.The results of orthogonal experimental variance analysis showed that there was no significant difference in the phenol content among the three factors(particle size,liquid-to-solid ratio,and soaking time)within the set gradient of the experiment(P>0.05,α=0.05);based on the results of orthogonal experiments and comprehensive consideration of industrial production energy consumption,the optimal process conditions for water vapor distillation of eugenol were determined as:particle size 80 mesh,liquid-to-solid ratio 1∶14(g/mL),and soaking time 6 h;the optimal process conditions for bancroftinone were:particle size 80 mesh,liquid-to-solid ratio 1∶12(g/mL),and soaking time 24 h.[Conclusion]The verification test confirmed that under the above optimized process conditions,the amount of target effective components in Eugenia caryophyllata Thunb.and Syzygium aromaticum(L.)Merr.&L.M.Perry extracts was the highest.This process is stable and has good repeatability,and can be used as a standardized industrial process for efficient extraction of eugenol and bancroftinone.
