不同产区赤霞珠葡萄根际微生物特征与葡萄果实品质相关性研究OA
A Study on Correlation between Rhizosphere Microbial Characteristics and Berry Quality of Cabernet Sauvignon in Different Producing Areas
[目的]明确贺兰山东麓酿酒葡萄(赤霞珠)根际土壤微生物群落特征与浆果品质的关联机制,完善产区微生物风土调控理论.[方法]以贺兰山东麓产区7个代表性酒庄10年生赤霞珠为研究对象,系统采集根际土壤及成熟浆果样品.基于16S rRNA和ITS rRNA高通量测序技术,解析土壤细菌、真菌群落结构与多样性,同步测定浆果可溶性固形物、可滴定酸、单宁、总酚及花色苷等核心品质指标,并通过Pearson相关性分析揭示二者耦合关系.[结果]不同酒庄赤霞珠根际微生物α多样性指数(Chao1、Shannon)存在显著差异(P<0.05),美贺酒庄(M1)细菌多样性最高,玉泉营酒庄(M4)最低.浆果品质方面,M1产区可溶性固形物(29.67 °Brix)和花色苷(5.00 mg/g)含量显著高于其他酒庄,M4可溶性固形物(24.50 °Brix)含量最低;可滴定酸含量为5.15~6.59 g/L,其中立兰酒庄(M3)最低.土壤芽孢杆菌属、假单胞菌属等有益菌丰度与浆果可溶性固形物、总酚及花色苷含量呈显著正相关(P<0.05),真菌类群丰度与可滴定酸含量呈负相关;细菌门水平中,芽单胞菌门(Gemmatimonadota)、脱硫杆菌门(Desulfobacterota)等与品质指标呈正相关,疣微菌门(Verrucomicrobiota)、拟杆菌门(Bacteroidota)等与品质指标呈负相关;真菌门水平中,子囊菌门(Ascomycota)、壶菌门(Chytridiomycota)与品质指标呈正相关,球囊菌门(Glomeromycota)、被孢霉门(Mmortierellomycota)与品质指标呈负相关.[结论]贺兰山东麓赤霞珠根际微生物群落结构具有显著的产区特异性,可通过调控土壤养分循环介导浆果品质分异.
[Objective]To clarify the association mechanism between the microbial community characteristics in the rhizosphere soil of wine grapes(Cabernet Sauvignon)on the eastern foothills of Helan Mountain and the quality of the berries,and to improve the theory of microbial terroir regulation in the production area.[Method]With 10-year-old Cabernet Sauvignon grapes from 7 representative wineries in the Helan Mountain production area as the research objects,and samples of rhizosphere soil and mature berries were systematically collected.Based on high-throughput sequencing technologies of 16S rRNA and ITS rRNA,the structure and diversity of soil bacterial and fungal communities were analyzed,and core quality indicators such as soluble solids,titratable acid,tannin,total phenols,and anthocyanins were simultaneously determined.Pearson correlation analysis was used to reveal the coupling relationship between the two.[Result]There were significant differences in the α-diversity index(Chao1,Shannon)of rhizosphere microorganisms among different wineries(P<0.05).The bacterial diversity was the highest in Meihe Winery(M1),and the lowest in Yuquanying Winery(M4).In terms of berry quality,the soluble solids(29.67 °Brix)and anthocyanin(5.00 mg/g)content in M1 production area were significantly higher than those in other wineries,while the soluble solids content(24.50 °Brix)was the lowest in M4.The titratable acid content generally fluctuated within the range of 5.15-6.59 g/L,with the lowest content in Lilan Winery(M3).The abundance of beneficial bacteria such as Bacillus,Pseudomonas,etc.was significantly positively correlated with the soluble solids,total phenols,and anthocyanin contents of the berries(P<0.05),while the abundance of fungal groups was negatively correlated with the titratable acid content.At the bacterial phylum level,the Gemmatimonadota,Desulfobacterota,etc.were positively correlated with the quality indicators,while the Verrucomicrobiota,Bacteroidota,etc.were negatively correlated with the quality indicators;at the fungal phylum level,the Ascomycota,Chytridiomycota,etc.were positively correlated with the quality indicators,while the Glomeromycota,Mortierellomycota were negatively correlated with the quality indicators.[Conclusion]The microbial community structure in the rhizosphere of Cabernet Sauvignon on the eastern foothills of Helan Mountain has significant regional specificity,and it can be mediated by regulating soil nutrient cycling to cause berry quality differentiation.The research results can provide support for the exploration of terroir potential in the production area and the improvement of wine quality differentiation.
