"引阳入阴"法针刺对失眠模型大鼠海马Nek7/NLRP3/Caspase-1/IL-1β信号通路的影响OA
Effect of Acupuncture with the Method of"Inducing Yang into Yin"on Nek7/NLRP3/Caspase-1/IL-1β Signal Pathway in Hippocampus of Insomnia Model Rats
目的 观察"引阳入阴"法针刺对失眠模型大鼠海马NIMA相关蛋白激酶7/核苷酸结合寡聚化结构域样受体蛋白3/半胱氨酸天冬氨酸蛋白水解酶-1/白细胞介素-1β(Nek7/NLRP3/Caspase-1/IL-1β)信号通路的影响,探讨针刺改善失眠的作用机制.方法 将40只SPF级健康雄性SD大鼠随机分为空白组10只,造模组30只.造模组采用腹腔注射氯苯丙氨酸(P-chlorophenylalanine,PCPA)的方法构建失眠模型.随机选取造模成功的30只大鼠,分为模型组、针刺组、西药组,每组10只.针刺组给予"引阳入阴"法针刺,穴取百会、外关(双)、内关(双)、胃俞(双)、中脘、太溪(双),每日1次,连续7 d;西药组用艾司唑仑溶液灌胃,每日1次,连续7 d.通过戊巴比妥钠翻正实验和旷场实验评价各组大鼠失眠行为;利用苏木素-伊红(HE)染色法观察大鼠海马组织病理改变;ELISA法检测大鼠海马IL-1β、白细胞介素-18(IL-18)含量;用RT-qPCR法检测大鼠海马Nek7、NLRP3、凋亡相关斑点样蛋白(ASC)、Caspase-1、IL-1β、IL-18 mRNA表达;用Western blot法检测海马中Nek7、NLRP3、ASC、Caspase-1、IL-1β、IL-18蛋白表达.结果 与空白组比较,造模大鼠体质量明显下降(P<0.01),睡眠潜伏期时长明显延长,睡眠时间明显缩短(P<0.01),旷场实验中运动总距离和运动时间明显增多(P<0.01),中心区域停留时间减少(P<0.05),海马区组织萎缩,神经元数量明显减少并伴排列紊乱,部分细胞核固缩变形,胞质浓缩深染,局部区域出现点状坏死灶及微量出血现象,大鼠海马IL-1β、IL-18含量明显升高(P<0.01),Nek7、NLRP3、ASC、Caspase-1、IL-1β和IL-18 的mRNA及蛋白表达水平均显著升高(P<0.01).与模型组相比,西药组和针刺组体质量均明显增加(P<0.05,P<0.01),睡眠潜伏期缩短(P<0.01),睡眠时间明显延长(P<0.01),运动总距离和运动时间明显减少(P<0.01),中心区域停留时间明显增加(P<0.05,P<0.01),神经元数量接近正常水平,细胞排列基本有序,核仁清晰可见,仅偶见点状坏死灶及个别胞质浓缩现象,海马IL-1β、IL-18含量显著降低(P<0.01),Nek7、NLRP3、ASC、Caspase-1、IL-1β、IL-18 mRNA及蛋白表达水平均不同程度降低(P<0.05,P<0.01).与西药组相比,针刺组运动总距离和运动时间明显减少(P<0.05),ASC、Caspase-1、IL-1β、IL-18 mRNA表达降低(P<0.05,P<0.01),Nek7、NLRP3、ASC、Caspase-1、IL-1β、IL-18蛋白表达水平均降低(P<0.05,P<0.01).结论 "引阳入阴"法针刺可改善失眠大鼠的睡眠质量,减轻海马神经炎症反应,其机制可能与调节Nek7/NLRP3/Caspase-1/IL-1β信号通路有关.
Objective To observe the effect of acupuncture with the method of"inducing Yang into Yin"on the signal pathway of NIMA-related protein kinase 7/oligonucleotide-binding domain-like receptor protein 3/cysteine aspartate proteolytic enzyme-1/interleukin-1β(Nek7/NLRP3/Caspase-1/IL-1β)in hippocampus of insomnia model rats,and to explore the mechanism of acupuncture in improving insomnia.Methods Forty SPF healthy male SD rats were randomly divided into blank group(10 rats)and model group(30 rats).The insomnia model was established by intraperitoneal injection of p-chlorophenylalanine(PCPA).Randomly select 30 rats that successfully established the model and divide them into model group,acupuncture group and Western medicine group,with 10 rats in each group.The acupuncture group was given acupuncture by the method of"inducing Yang into Yin",and the points were"Baihui(GV20)""Waiguan(TE5)(double)""Neiguan(PC6)(double)""Weishu(BL21)(double)""Zhongwan(CV12)"and"Taixi(KI3)(double)"once a day for 7 days.The Western medicine group was given estazolam solution once a day for 7 days.The insomnia behavior of rats in each group was evaluated by pentobarbital sodium correction test and open field test.HE matoxylin-eosin staining was used to observe the pathological changes of hippocampus in rats.The contents of IL-1β and IL-18 in hippocampus of rats were detected by ELISA.The mRNA expressions of Nek7,NLRP3,apoptosis-related spot-like protein(ASC),Caspase-1,IL-1β and IL-18 were detected by RT-qPCR.The expressions of Nek7,NLRP3,ASC,Caspase-1,IL-1β and IL-18 in hippocampus were detected by Western blot.Results Compared with the blank group,the weight of rats in each group decreased significantly(P<0.01).The duration of sleep latency was significantly prolonged and the sleep time was significantly shortened(P<0.01).In the open field experiment,the total distance and time of exercise increased obviously(P<0.01),and the residence time in the central area decreased(P<0.05).The hippocampus tissue atrophied,the number of neurons decreased obviously with disordered arrangement,some nuclei contracted and deformed,cytoplasm concentrated and stained deeply,and punctate necrotic foci and micro-bleeding appeared in some areas.The contents of IL-1β and IL-18 in hippocampus of rats increased significantly(P<0.01).The mRNA and protein expression levels of Nek7,NLRP3,ASC,Caspase-1,IL-1β and IL-18 all increased significantly(P<0.01).Compared with the model group,the body weight of the Western medicine group and the acupuncture group increased significantly(P<0.05,P<0.01).The sleep latency was shortened(P<0.01)and the sleep time was obviously prolonged(P<0.01).The total distance and time of exercise decreased significantly(P<0.01),and the stay time in the central area increased significantly(P<0.05,P<0.01).The number of neurons is close to the normal level,the cell arrangement is basically orderly,the nucleoli is clearly visible,and only punctate necrotic foci and individual cytoplasm concentration are occasionally seen.The contents of IL-1β and IL-18 in hippocampus decreased significantly(P<0.01).The mRNA and protein expression levels of Nek7,NLRP3,ASC,Caspase-1,IL-1β,IL-18 decreased in different degrees(P<0.05,P<0.01).Compared with Western medicine group,the total distance and time of exercise in acupuncture group were significantly reduced(P<0.05).The expression of ASC,Caspase-1,IL-1β and IL-18 mRNA decreased(P<0.05,P<0.01).The expression levels of Nek7,NLRP3,ASC,Caspase-1,IL-1β and IL-18 decreased(P<0.05,P<0.01).Conclusion Acupuncture with the method of"inducing Yang into Yin"can improve the sleep quality of insomnia rats and reduce the inflammatory reaction of hippocampus,and its mechanism may be related to the regulation of Nek7/NLRP3/caspase-1/IL-1β signal pathway.
