雷公藤甲素通过周细胞EMT-like通路改善百草枯诱导的小鼠肺纤维化及其作用机制OA
Analysis on Triptolide Improving Paraquat-induced Pulmonary Fibrosis in Mice through Pericellular EMT-like Pathway and Its Mechanism
目的 探讨雷公藤甲素通过周细胞(pericyte)对百草枯诱导的小鼠肺纤维化的改善作用及机制.方法 选60只C57BL/6雄性小鼠,随机分为5组,每组12只,即空白对照组、模型组、雷公藤甲素低剂量组、雷公藤甲素中剂量组、雷公藤甲素高剂量组,用百草枯造模构建肺纤维化模型,研究药物对其体质量、肺系数、肺组织胶原含量的影响,按炎性细胞浸润、肺间质纤维化程度等指标对肺组织切片评分,测定羟脯氨酸(Hyp)含量,检测血清还原型谷胱甘肽(GSH)、一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)及炎症因子等指标水平,通过免疫组化及Western blot技术系统评估雷公藤甲素对上皮-间质转化(EMT)标志物的调控作用,观察转化生长因子-β1(TGF-β1)、Smad3 mRNA及蛋白相对表达量的变化情况.结果 模型组小鼠肺组织Hyp含量较对照组显著升高(P<0.01);雷公藤甲素低、中、高剂量组Hyp含量分别较模型组降低(P<0.05);与雷公藤甲素低、中剂量组比较,雷公藤甲素高剂量组小鼠肺组织中Hyp含量降低(P<0.05).各组小鼠血清GSH、NO、SOD和MDA指标水平有明显差异;与对照组比较,模型组小鼠肺组织中IL-1β、IL-18和TNF-α表达水平显著升高(P<0.01);与模型组比较,雷公藤甲素低、中、高剂量组肺组织中IL-1β、IL-18和TNF-α表达水平均有显著下降(P<0.05);与雷公藤甲素低、中剂量组比较,雷公藤甲素高剂量组IL-1β、IL-18和TNF-α表达水平显著下降(P<0.05);与对照组比较,模型组小鼠肺泡微血管周细胞中TGF-β1、Smad3 的mRNA和蛋白表达水平均升高(P<0.01);雷公藤甲素低、中、高剂量组小鼠肺泡微血管周细胞中TGF-β1、Smad3 的mRNA和蛋白表达水平均低于模型组(P<0.05),且与雷公藤甲素低、中剂量组比较,雷公藤甲素高剂量组小鼠肺泡微血管周细胞中TGF-β1、Smad3 的mRNA和蛋白表达水平均降低(P<0.05).结论 该研究证明了雷公藤甲素通过调控周细胞EMT-like通路,靶向TGF-β1发挥抗肺纤维化的作用.
Objective To explore the improvement effect and mechanism of triptolide on paraquat-induced pulmonary fibrosis in mice through pericytes.Methods Sixty male C57BL/6 mice were randomly assigned to five groups,with 12 mice in each group:a blank control group,a model group,a low-dose triptolide group,a medium-dose triptolide group,and a high-dose triptolide group.A pulmonary fibrosis model was established using paraquat.The effects of the drug on body weight,lung coefficient,and collagen content in lung tissue were investigated.Lung tissue sections were scored based on indicators such as inflammatory cell infiltration and the degree of pulmonary interstitial fibrosis.The content of hydroxyproline(Hyp)was measured,and changes in serum levels of reduced glutathione(GSH),nitric oxide(NO),superoxide dismutase(SOD),malondialdehyde(MDA),and inflammatory factors were detected.Immunohistochemistry and Western blot techniques were used to systematically evaluate the regulatory effect of triptolide on epithelial-mesenchymal transition(EMT)markers,and changes in the relative expression levels of TGF-β1,Smad homolog 3(Smad3)mRNA and protein were observed.Results The content of Hyp in the lung tissue of mice in the model group was significantly higher than that in the control group(P<0.01).The Hyp content in the low,medium,and high-dose triptolide groups were reduced compared to the model group(P<0.05).Compared with the low and medium-dose triptolide groups,the Hyp content in the lung tissue of mice in the high-dose triptolide group was further decreased(P<0.05).Significant differences were observed in the levels of serum GSH,NO,SOD,and MDA among the groups of mice.Compared to the control group,the expression levels of IL-1β,IL-18,and TNF-α in the lung tissue of mice in the model group were significantly increased(P<0.01).Compared to the model group,the expression levels of IL-1β,IL-18,and TNF-α in the lung tissue of mice in the low,medium,and high-dose triptolide groups were significantly decreased(P<0.05).Compared to the low and medium-dose triptolide groups,the expression levels of IL-1β,IL-18,and TNF-α in the lung tissue of mice in the high-dose triptolide group were further decreased(P<0.05).Compared to the control group,the mRNA and protein expression levels of TGF-β1 and Smad3 in the alveolar microvascular pericytes of mice in the model group were increased(P<0.01).The mRNA and protein expression levels of TGF-β1 and Smad3 in the alveolar microvascular pericytes of mice in the low,medium,and high-dose triptolide groups were lower than those in the model group(P<0.05).Furthermore,compared to the low and medium-dose triptolide groups,the mRNA and protein expression levels of TGF-β1 and Smad3 in the alveolar microvascular pericytes of mice in the high-dose triptolide group were further decreased(P<0.05).Conclusion This study demonstrated that triptolide regulates the pericyte EMT-like pathway and targets TGF-β1 to play an anti-pulmonary fibrosis role.
