弥漫大B细胞淋巴瘤中SUMO2的表达及与预后的相关性分析OA

Objective To investigate SUMO2 expression in diffuse large B-cell lymphoma(DLBCL)and its asso-ciation with prognosis.Methods Bioinformatics analyses based on the GEPIA2 and GEO public databases were per-formed to compare SUMO2 expression between DLBCL tissues and control tissues with reactive lymphoid hyperplasia and to evaluate the association between SUMO2 expression and overall survival.Paraffin-embedded tissue samples from 145 patients with DLBCL and 43 controls with reactive lymphoid hyperplasia were collected.SUMO2 protein ex-pression was detected by immunohistochemistry,and its associations with clinicopathological characteristics and prog-nosis were analyzed.Univariate and multivariate Cox regression models were used to identify risk factors for poor prog-nosis in patients with DLBCL.Results GEPIA2 data showed that SUMO2 mRNA expression was significantly higher in DLBCL tissues(median,302.375)than in control tissues(median,42.739,P<0.01).The rate of high SUMO2 protein expression was 63.44%(92/145)in DLBCL tissues and 32.56%(14/43)in control tissues.The SUMO2 H-score was significantly higher in the DLBCL group than in the control group(t=4.619,P<0.001).In the GEO data-base,SUMO2 mRNA expression was significantly associated with Hans subtype,lactate dehydrogenase level,and prognosis(P<0.05).In the clinical samples,SUMO2 protein expression was significantly associated with BCL2 gene,MYC protein expression,MYC/BCL2 protein coexpression,and prognosis(P<0.05).Overall survival was sig-nificantly shorter in patients with high SUMO2 mRNA expression than in those with low expression in the GEO data-base(P<0.01).Similarly,overall survival was significantly shorter in patients with high SUMO2 protein expression than in those with low expression in the clinical cohort(P<0.01).Univariate and multivariate Cox regression analy-ses showed that high SUMO2 expression was an independent risk factor for poor prognosis in patients with DLBCL(HR=2.310,95%CI:1.347-3.961,P<0.01).Conclusion SUMO2 expression is upregulated in DLBCL tis-sues,and high SUMO2 expression is an independent risk factor for poor prognosis in patients with DLBCL.
