LncRNA PVT1/c-Myc表达对弥漫大B细胞淋巴瘤的影响及相关机制分析OA
Effects of lncRNA PVT1/c-Myc expression on diffuse large B-cell lymphoma and the underlying mechanisms
目的 探讨弥漫大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)中PVT1对c-Myc表达和磷酸化的影响及其与肿瘤发生、发展的关系.方法 采用免疫组化法和RT-qPCR技术检测DLBCL和反应性增生淋巴结(reactive hyperplastic lymph nodes,RHLN)中PVT1、c-Myc、Ki67等表达,并分析PVT1与c-Myc表达的相关性.应用RT-qPCR、Western blot法检测DLBCL细胞模型中PVT1、c-Myc、PCNA、p65、c-Myc(phospho T58)、c-Myc(phospho S62)mRNA或蛋白表达.运用CCK-8及流式细胞术检测PVT1异常表达对DLBCL细胞增殖、周期及凋亡的影响.结果 与RHLN相比,PVT1、c-Myc在DLBCL中的表达增高且两者呈正相关(P<0.001,r=0.782),与患者临床分期、Ki67表达相关(P<0.05).过表达PVT1时DLBCL中c-Myc、p65及PCNA表达明显升高,反之亦然;过表达PVT1时c-Myc(phospho T58)蛋白表达明显降低,敲低则出现相反趋势(P<0.05).结论 DLBCL中PVT1和c-Myc表达呈正相关,两者过表达与DLBCL不良预后和肿瘤进展相关.PVT1可能通过上调c-Myc的表达促进DLBCL发生、发展,提示PVT1可能是DLBCL治疗和预后评估的潜在靶点.
Objective To investigate the effects of PVT1 on c-Myc expression and phosphorylation in diffuse large B-cell lymphoma(DLBCL)and its role in tumorigenesis and progression.Methods Immunohistochemistry and RT-qPCR were used to evaluate PVT1,c-Myc,and Ki67 expression in DLBCL tissues and reactive hyperplastic lymph nodes(RHLNs),and the correlation between PVT1 and c-Myc expression was analyzed.RT-qPCR and Western blot-ting were used to measure the mRNA or protein expression of PVT1,c-Myc,PCNA,p65,phospho-c-Myc(Thr58),and phospho-c-Myc(Ser62)in DLBCL cell models.CCK-8 assays and flow cytometry were used to determine the ef-fects of aberrant PVT1 expression on DLBCL cell proliferation,cell-cycle progression,and apoptosis.Results PVT1 and c-Myc expression levels were significantly higher in DLBCL tissues than in RHLNs,and their expression levels were positively correlated(P<0.001,r=0.782).Both were associated with clinical stage and Ki67 expression(P<0.05).PVT1 overexpression significantly increased c-Myc,p65,and PCNA expression in DLBCL cells,whereas PVT1 knockdown had the opposite effects.PVT1 overexpression significantly decreased phospho-c-Myc(Thr58)protein expression,whereas PVT1 knockdown produced the opposite trend(P<0.05).Conclusion PVT1 and c-Myc expression levels are positively correlated in DLBCL,and their overexpression is associated with poor prog-nosis and tumor progression.PVT1 may promote DLBCL tumorigenesis and progression by upregulating c-Myc expres-sion,suggesting that PVT1 may be a potential target for DLBCL treatment and prognostic assessment.
