牛至精油对羔羊盲肠组织形态、菌群结构及功能的影响OA
Effect of Oregano Essential Oil on Cecal Tissue Morphology,Microbial Community Structure and Function in Lambs
试验旨在筛选出萨能奶山羊羔羊科学合理的牛至精油(OEO)添加水平.选择出生体重相近的48只萨能奶山羊公羔,随机分为3个处理组(每组4个重复,每个重复4只羔羊),羔羊7日龄开始饲喂添加0 g/d(CON)、1 g/d(OEO1)和2 g/d(OEO2)OEO的开食料.70日龄清晨空腹称重,各组随机屠宰6只羔羊,采集盲肠组织和内容物进行检测.结果表明:OEO对羔羊盲肠pH和组织形态无显著影响(P>0.05).18个盲肠内容物样品共得到1522540 条有效数据和 5157 个 ASV,OEO2 组 chao1、shannon、simpson、observed_features和pielou_e指数极显著高于OEO1组和CON组(P<0.01),dominance指数CON组极显著高于OEO1组和OEO2组(P<0.01);LEfSe分析发现,OEO1组主要是颤螺菌目(Oscil-lospirales)的颤螺菌科(Oscillospiraceae);OEO2组显著富集的有芽孢杆菌门(Bacillota)的克里斯滕森氏菌(Christensenellaceae_R-7_group)和单球菌属(Monoglobus),拟杆菌门(Bacteroidota)的拟杆菌属(Bacteroides)、普雷沃氏菌科 UCG-004(Prevotellaceae UCG-004)和理研菌科_RC9肠群(Rikenellaceae_RC9_gut-group);CON组中显著富集的包括Bacillo-ta 的琼脂杆菌属(Agathobacter)、粪球菌属(Coprococcus)和瘤胃球菌属(Ruminococcus).运用Tax4Fun法预测分析得出羔羊盲肠段内容物微生物以碳水化合物代谢、膜转运、复制与修复、氨基酸代谢、翻译、能量代谢、核苷酸代谢、信号转导、细胞运动性、辅助因子和维生素代谢为主.相关性分析结果显示,pH 与 Agathobacter 和 unclassified_Clostridia_UCG.014 之间呈正相关关系(P<0.05),与 Christensenellaceae_R.7_group、unclassified_UCG.010 和Rikenellaceae_RC9_gut_group 之间呈负相关关系(P<0.05),相对容积与 Bacteroides 之间呈负相关关系(P<0.05),黏膜层厚与 Bacteroides、unclassified_.Eubacterium._coprostanoli-genes_group和Monoglobus之间呈负相关关系(P<0.05).综上所述,在羔羊生产中使用OEO可以有效改善盲肠微生物菌群结构及功能,且牛至精油的最适添加剂量为2 g/d.
This experiment aimed to screen the optimal supplementation level of oregano essential oil(OEO)for Saanen goat kids.Forty-eight male Saanen goat kids with similar birth weights were selected and randomly assigned to three treatment groups(4 replicates per group,4 kids per replicate).At 7 days of age,the kids were fed starter diets supplemented with 0 g/d(CON),1 g/d(OEO1),and 2 g/d(OEO2)OEO.At 70 days of age,the kids were weighed in the morning after fasting,and 6 kids from each group were randomly slaughtered to collect cecal tissue and contents for analysis.The results showed that OEO had no significant effects on cecal pH or tissue morphology(P>0.05).A total of 1,522,540 high-quality reads and 5,157 ASVs were obtained from the 18 cecal content samples.The chao1,shannon,simpson,observed_features,and pielou_e indices in the OEO2 group were extremely higher than those in the OEO1 and CON groups(P<0.01),while the dominance index in the CON group was significantly higher than that in the OEO1 and OEO2 groups(P<0.01).LEfSe analysis revealed that the OEO1 group was primarily characterized by Oscillospiraceae within the order Oscillospirales.The OEO2 group showed significant enrichment of Christensenellaceae_R-7_group and Monoglobus(phylum Bacillota),as well as Bacteroides,Prevotellaceae UCG-004,and Rikenellaceae_RC9_gut_group(phylum Bacteroidota).The CON group showed significant enrichment of Agathobacter,Coprococcus,and Ruminococcus(phylum Ba-cillota).Tax4Fun functional prediction analysis indicated that the cecal microbiota were primarily involved in carbohydrate metabolism,membrane transport,replication and repair,amino acid metabolism,transla-tion,energy metabolism,nucleotide metabolism,signal transduction,cell motility,and metabolism of co-factors and vitamins.Correlation analysis showed that pH was positively correlated with Agathobacter and unclassified_Clostridia_UCG.014(P<0.05),and negatively correlated with Christensenellaceae_R.7_group,unclassified_UCG.010,and Rikenellaceae_RC9_gut_group(P<0.05).Relative cecal volume was negatively correlated with Bacteroides(P<0.05),and mucosal layer thickness was negatively correla-ted with Bacteroides,unclassified_Eubacterium_coprostanoligenes_group,and Monoglobus(P<0.05).In conclusion,the use of OEO in goat kid production can effectively improve the structure and function of the cecal microbiota,and the optimal dietary supplementation level of oregano essential oil is 2 g/d.
