过瘤胃蛋氨酸对藏西北白绒山羊部分消化道酶活及瘤胃和空肠微生物菌群的影响OA
Effects of Rumen-Protected Methionine on Partial Digestive Enzyme Activities and the Microbiota of Rumen and Jejunum in Northwest Xizang Cashmere Goats
试验旨在探究过瘤胃蛋氨酸对藏西北白绒山羊部分消化道酶活及瘤胃和空肠微生物菌群的影响.选取体况健康的12月龄藏西北白绒山羊16只,随机分为2组(公母各半),每组8个重复,每个重复1只羊.对照组(CON组)饲喂基础日粮,试验组(RPMet组)在基础日粮中添加0.5%过瘤胃蛋氨酸.预试期1周,正试期7周.结果表明:与CON组相比:(1)RPMet组瘤胃蛋白酶活性极显著升高(P<0.01),空肠和盲肠胰蛋白酶活性极显著升高(P<0.01),结肠和盲肠糜蛋白酶活性极显著升高(P<0.01).(2)RPMet组瘤胃菌群Chao1指数显著升高(P<0.05).(3)门水平中,RPMet组瘤胃菌群厚壁菌门(Firmicutes)相对丰度极显著升高(P<0.01);属水平中,RPMet组瘤胃菌群普氏菌属(Prevotella)、瘤胃球菌属(Rumi-nococcus)与琥珀酸菌(Succiniclasticum)相对丰度极显著升高(P<0.01).(4)缬氨酸、亮氨酸和异亮氨酸的降解通路极显著上调(P<0.01),生物素和色氨酸代谢通路显著上调(P<0.05).综上,饲粮补充过瘤胃蛋氨酸提高了白绒山羊部分消化道蛋白酶活性,改变了白绒山羊的瘤胃菌群结构,对白绒山羊瘤胃菌群氨基酸代谢产生了影响,在一定程度上提高了白绒山羊瘤胃菌群对饲粮纤维的消化能力.
This experiment aimed to investigate the effects of rumen-protected methionine on the activ-ities of certain digestive enzymes and the microbial communities in the rumen and jejunum of Northwest Xizang cashmere goats.Sixteen healthy 12-month-old Northwest Xizang white cashmere goats(males and females equally divided)were randomly assigned into two groups with eight replicates per group and one goat per replicate.The control group(CON)was fed a basal diet,while the experimental group(RPMet)was fed the basal diet supplemented with 0.5%rumen-protected methionine.The adaptation period lasted one week,followed by a seven-week experimental period.The results showed that compared with the CON group:(1)rumen pepsin activity in the RPMet group was extremely increased(P<0.01);jejunal and ce-cal trypsin activities were also extremely increased(P<0.01);and colonic and cecal chymotrypsin activi-ties were extremely increased(P<0.01).(2)The Chao1 index of the rumen microbiota in the RPMet group was increased(P<0.05).(3)At the phylum level,the relative abundance of Firmicutes in the ru-men microbiota of the RPMet group was extremely increased(P<0.01);at the genus level,the relative a-bundances of Prevotella,Ruminococcus,and Succiniclasticum in the rumen microbiota of the RPMet group were significantly increased(P<0.01).(4)The degradation pathways of valine,leucine,and iso-leucine were extremely upregulated(P<0.01),while the biosynthetic pathways of biotin and tryptophan metabolism were significantly upregulated(P<0.05).In conclusion,dietary supplementation with rumen-protected methionine increased the activities of certain digestive proteases,altered the rumen microbial community structure,and influenced amino acid metabolism of the rumen microbiota in cashmere goats,thereby enhancing,to a certain extent,the digestive capacity of the rumen microbiota for dietary fiber.
