目的:建立拳参标准汤剂薄层色谱(TLC)和定量特征图谱方法,为其与混伪品的鉴别及质量控制提供依据.方法:采用TLC法,以拳参对照药材、没食子酸、绿原酸为对照,对拳参标准汤剂、配方颗粒及其易混品进行鉴别;同时采用超高效液相色谱(UPLC)法建立拳参定量特征图谱,结合相似度软件、聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)法确定拳参与其混伪品的主要差异成分并定量测定.结果:TLC图结果显示,拳参标准汤剂与其易混品在荧光斑点上存在明显差异.通过相似度评价软件分析15批拳参标准汤剂特征图谱,建立对照图谱,拳参样品的相似度>0.90,而珠芽蓼和草血竭标准汤剂的相似度均<0.90.CA、PCA、OPLS-DA表明,拳参与其混伪品的主要差异成分为没食子酸、绿原酸,定量结果显示两者含量存在明显差异.结论:本研究建立的TLC和HPLC法能够快速、准确、客观地对拳参与其易混品进行鉴别,显著地提高了临床用药的准确性.

Objective:To establish methods for thin-layer chromatography(TLC)and quantitative characteristic chromatogram to provide a scientific basis for the identification and quality control of Quanshen(Polygonum Bistorta L)standard decoction and its adulterants.Methods:TLC was employed to identify the Quanshen(Polygonum Bistorta L.)standard decoction,formula granules,and its adulterants,using Quanshen(Polygonum Bistorta L.)reference herbs,gallic acid and chlorogenic acid as controls.Additionally,ultra-performance liquid chromatog-raphy(UPLC)was used to establish a quantitative characteristic chromatogram of Quanshen(Polygonum Bistorta L.).The characteristic chromatograms were analyzed using similarity evaluation software,cluster analysis(CA),principal component analysis(PCA)and orthogonal partial least square discriminant analysis(OPLS-DA)to identify the main differences between Quanshen(Polygonum Bistorta L.)and its adulterants.The differential components were quantitatively determined.Results:TLC revealed distinct fluorescence spots between the standard decoction of Quanshen(Polygonum Bistorta L)and its adulterants.The similarity evaluation software analyzed the characteristic chromatograms of 15 batches of Quanshen(Polygonum Bistorta L.)standard decoction,establishing a reference chromatogram.The similarity of Quanshen(Polygonum Bistorta L.)samples was greater than 0.90,while the similarity of adulterants(such as Polygonum viviparum L.and Polygonum paleaceum Wall.)was less than 0.90.CA,PCA and PLS-DA identified gallic acid and chlorogenic acid as the primary differential components between Quanshen(Polygonum Bistorta L.)and its adulterants.Quantitative results showed significant differences in the content of these two components.Conclusion:The established TLC and liquid chromatography methods can quickly,accurately and objectively identify Quanshen(Polygonum Bistorta L.)and its adulterants,significantly improving the accuracy of clinical drug use.
