石榴皮多酚软膏调节GATA6及相关因子抗痤疮机制OA
Anti-Acne Mechanism of Pomegranate Peel Polyphenols Ointment via Regulation of GATA6 and Related Factors
目的:通过观察不同浓度石榴皮多酚软膏对金黄地鼠皮脂腺斑及血清中转录因子GATA6、转化生长因子-β(TGF-β)、白细胞介素-10(IL-10)表达的影响,探讨石榴皮多酚软膏调节GATA6抗痤疮的作用及其机制.方法:将36只金黄地鼠随机分为6组[空白组、基质组(1 g软膏基质)、低浓度组(0.480%石榴皮多酚软膏)、中浓度组(0.960%石榴皮多酚软膏)、高浓度组(1.920%石榴皮多酚软膏)及阳性对照组(0.025%异维A酸乳膏)],连续给药4周后观察药物干预效果.HE染色观察皮脂腺斑组织病理学改变情况;酶联免疫吸附试验(ELISA)检测血清IL-10、TGF-β1、TGF-β2水平;蛋白质印迹法(Western blotting)检测皮脂腺斑GATA6表达水平.结果:HE染色光学显示,各浓度石榴皮多酚软膏可剂量依赖性缩小皮脂腺腺体,减少腺叶与成熟皮脂腺细胞,高浓度组改善效果与阳性对照接近.基质组金黄地鼠血清IL-10水平低于空白组(P<0.05),TGF-β1、TGF-β2水平高于空白组(P<0.05);各给药组均可逆转基质诱导的因子紊乱,上调IL-10,下调TGF-β1、TGF-β2(P<0.05),整体呈浓度依赖效应,高浓度组作用最优,低、中浓度组疗效温和且组间差异无统计学意义(P>0.05).基质组金黄地鼠皮脂腺斑GATA6表达低于空白组(P<0.05);各给药组GATA6表达均显著升高(P<0.05),高浓度组与阳性对照组上调幅度相当(P>0.05).结论:石榴皮多酚软膏可能通过调控GATA6及其相关因子,影响炎性因子分泌、防角化异常等,从而抗痤疮形成.
Objective:To investigate the anti-acne mechanism of pomegranate peel polyphenols ointment by observing their effects on sebaceous gland spots and serum levels of transcription factor GATA6,transforming growth factor-β(TGF-β),and interleukin-10(IL-10)in golden hamsters.Methods:Totally 36 golden hamsters were randomly divided into six groups[blank group,vehicle group(1 g ointment base),low-dose group(0.480%pomegranate peel polyphenols ointment),medium-dose group(0.960%pomegranate peel polyphenols ointment),high-dose group(1.920%pomegranate peel polyphenols ointment),and positive control group(0.025%Isotretinol yogurt cream)].After 4 weeks of treatment,therapeutic effects were evaluated.Histopathological changes in sebaceous glands were assessed by Hematoxylin-Eosin(HE)staining.Serum IL-10,TGF-β,and TGF-β2 were quantified by enzyme linked immunosorbent assay(ELISA),and cutaneous GATA6 expression was measured by Western blotting.Results:HE staining results showed that pomegranate peel polyphenols ointment reduced sebaceous gland volume,glandular lobes and mature sebocytes in a dose-dependent manner,and the efficacy of high-dose group was similar to positive group.The vehicle group showed lower IL-10 level than blank group,while higher TGF-β1 and TGF-β2 level than blank group(P<0.05).All treatment groups reversed the above abnormal indexes,increased IL-10 and decreased TGF-β1 and TGF-β2(P<0.05),with obvious dose-dependent effect.The high-dose group presented the best therapeutic effect,while low-dose group and medium-dose group showed mild efficacy without significant inter-group difference(P>0.05).The GATA6 expression in vehicle group was lower than that of blank group(P<0.05).All treatment groups significantly up-regulated GATA6 expression(P<0.05),and there was no statistical difference between high-dose group and positive control group(P>0.05).Con-clusion:Pomegranate peel polyphenols ointment may exert its anti-acne effects by regulating GATA6 and its related factors,thereby influencing the secretion of inflammatory cytokines,preventing abnormal keratinization,and so on.
