目的:基于网络药理学与动物实验探讨骶尾部中药溻渍缓解肛肠疾病术后炎症的作用机制.方法:通过TCMSP数据库筛选溻渍方活性成分预测靶点,结合GeneCards、DisGeNET、OMIM、PharmGkb、TTD数据库获取术后相关靶点,构建药物-疾病网络,进行GO/KEGG富集分析.建立肛肠术后模型小鼠,检测脏器指数、组织病理、血清炎症因子hs-CRP、TNF-α、IL-6、IL-17,氧化应激指标T-AOC、MDA及MAPK通路蛋白表达.结果:网络药理学筛选出129种活性成分及148个交集靶点,核心成分包括槲皮素、大豆黄酮、芹菜素、山柰酚、大黄素,关键靶点涉及MAPK1、MAPK3、TNF、IL-6等,富集于MAPK、IL-17等通路.动物实验显示,溻渍方可显著降低模型小鼠炎症评分,降低血清hs-CRP、TNF-α、IL-6、IL-17及MDA水平,上调T-AOC表达水平,抑制ERK、p38蛋白表达,高剂量组效果最优(P<0.05).结论:骶尾部中药溻渍可能通过调控MAPK信号通路,降低hs-CRP、TNF-α、IL-6、IL-17等炎症因子的表达,发挥抗炎与抗氧化的双重作用,缓解肛肠术后炎症反应.

Objective:To investigate the mechanism of sacrococcygeal herbal application in reducing postoperative inflammation in anorectal surgery using network pharmacology and animal experiment.Methods:Active ingredients and targets were identified via TCMSP,while disease-related targets were obtained from GeneCards,DisGeNET,OMIM,PharmGkb,and TTD databases.A drug-disease network was constructed,and GO/KEGG enrichment analysis was performed.We established mouse models of post-anorectal surgery,and then measured organ indices,histopathological changes,serum inflammatory factors(hs-CRP,TNF—α,IL-6,IL-17),oxidative stress indicators(T-AOC,MDA),and the expression of MAPK pathway proteins.Results:Network analysis revealed 129 bioactive components and 148 therapeutic targets.The core active components comprise quercetin,daidzein,apigenin,kaempferol,and emodin.The key target genes include MAPK1,MAPK3,TNF,IL-6,and others,and these components are mainly enriched in signaling pathways such as the MAPK and IL-17 pathways.In vivo,the Tazifang(溻渍方)reduced inflammatory scores,decreased hs-CRP,TNF-α,IL-6,IL-17,MDA levels,enhanced T-AOC,and suppressed protein expression of ERK and p38.The high-dose group showed the best effect(P<0.05).Conclusion:The herbal application may alleviate postoperative inflammatory responses after anorectal surgery by regulating the MAPK signaling pathway,reducing the expression of inflam-matory cytokines such as hs-CRP,TNF-α,IL-6,and IL-17,thereby exerting dual anti-inflammatory and antioxidant effects.
