基于SLC7A11/GPX4通路探讨淫羊藿苷对绝经后骨质疏松症大鼠的影响OA
Effect of Icariin on Postmenopausal Osteoporosis Rats Based on SLC7A11/GPX4 Pathway
目的:探讨淫羊藿苷(ICA)调控溶质转运蛋白家族7成员11(SLC7A11)/谷胱甘肽过氧化物酶4(GPX4)轴改善去卵巢大鼠骨质疏松症的作用机制.方法:将36只8周龄健康雌性SD大鼠随机分为假手术(Sham)组、双侧卵巢切除(OVX)组、戊酸雌二醇(E2V)组、淫羊藿苷低剂量组(ICA-L组)、淫羊藿苷中剂量组(ICA-M组)、淫羊藿苷高剂量组(ICA-H组),每组6只.采用双侧卵巢摘除术建立绝经后骨质疏松大鼠模型(Sham组仅切除卵巢附近脂肪并实施腹部切口缝合操作).术后8周开始每日灌胃,Sham组与OVX组每日予生理盐水(10mL/kg),E2V组给予戊酸雌二醇(0.6 mg/kg),ICA-L组、ICA-M组、ICA-H组分别予低、中、高剂量ICA灌胃(50、100、200 mg/kg),持续8周.给药结束后,检测各组大鼠股骨骨微结构、骨密度(BMD)、相对骨体积(BV/TV)、骨小梁数量(Tb.N)、骨小梁分离度(Tb.Sp),酶联免疫吸附试验(ELISA)检测血清骨保护素(OPG)、核因子κB受体活化因子配体(RANKL)、抗酒石酸酸性磷酸酶5b(TRACP-5b)水平,试剂盒检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)、亚铁离子(Fe2+)水平,蛋白质印迹法(Western blotting)检测腰椎铁蛋白重链(FTH1)、SLC7A11、GPX4蛋白表达水平,实时荧定量PCR(qRT-PCR)法检测大鼠腰椎组织中SLC7A1 1 mRNA、GPX4 mRNA表达水平.结果:OVX组大鼠股骨呈现显著骨微结构退化,具体表现为骨小梁数量下降,伴随骨小梁网络排列紊乱,出现特征性的小梁结构萎缩及不规则形态学改变;ICA-L组大鼠骨小梁密度较OVX组略有改善;E2V组、ICA-M组、ICA-H组大鼠股骨组织骨小梁结构排列有序.OVX组大鼠BMD、BV/TV、Tb.N低于Sham组(P<0.01),Tb.Sp高于Sham组(P<0.01);E2V组、ICA-M、ICA-H 组大鼠 BMD、BV/TV、Tb.N 高于 OVX 组(P<0.05 或 P<0.01),Tb.Sp 低于 OVX 组(P<0.001);ICA-L 组大鼠 BMD 高于 OVX 组(P<0.05).OVX 组大鼠血清 OPG、OPG/RANKL、SOD 水平低于 Sham 组(P<0.01),RANKL、TRACP5b、MDA、Fe2+水平高于 Sham 组(P<0.01);E2V 组大鼠血清 OPG、OPG/RANKL、SOD 水平高于 OVX 组(P<0.05 或 P<0.01),RANKL、TRACP5b、MDA、Fe2+水平低于 OVX 组(P<0.01);ICA-H 组大鼠血清 OPG 水平高于 OVX组(P<0.01);ICA-L、ICA-M、ICA-H 组大鼠血清 SOD 水平、OPG/RANKL 高于 OVX 组(P<0.01),RANKL、TRACP5b、MDA、Fe2+水平低于OVX组(P<0.01).OVX组大鼠腰椎FTH1、SLC7A11、GPX4蛋白相对表达量低于Sham组(P<0.01);E2V组、ICA-H组大鼠腰椎FTH1、SLC7A11、GPX4蛋白相对表达量高于OVX组(P<0.01);ICA-M组大鼠腰椎SLC7A11、GPX4蛋白相对表达量高于OVX组(P<0.01);ICA-L组大鼠腰椎GPX4蛋白相对表达量高于OVX组(P<0.05).OVX组大鼠腰椎SLC7A11 mRNA、GPX4 mRNA相对表达量低于Sham组(P<0.01);E2V组、ICA-L组、ICA-M组、ICA-H组大鼠腰椎SLC7A11 mRNA、GPX4 mRNA相对表达量高于OVX组(P<0.01).结论:淫羊藿苷可通过SLC7A11/GPX4轴上调机制,降低OVX大鼠破骨细胞活性,抑制骨吸收,增加大鼠抗氧化能力并减轻脂质过氧化损伤,减少游离Fe2+蓄积,减少骨吸收,促进骨形成,修复骨小梁结构,发挥抗骨质疏松症作用.
Objective:To explore the mechanism of Icariin(ICA)regulating the axis of solute carrier family 7 member 11(SLC7A11)/glutathione peroxidase 4(GPX4)to improve osteoporosis in ovariectomized rats.Methods:Totally 36 8-week-old healthy female SD rats were randomly divided into Sham surgery(Sham)group,Bilateral ovariectomy(OVX)group,Estradiol valerate(E2V)group and Low-dose icariin group(ICA-L group),Middle-dose icariin group(ICA-M group)and High-dose icariin group(ICA-H group),with 6 rats in each group.The rat model of postmenopausal osteoporosis was established by bilateral ovariectomy(Sham group only removed the fat near the ovary and performed abdominal incision suture operation).Gastric lavage was given daily for 8 weeks after operation.Sham group and OVX group were given normal saline(10 mL/kg)daily.E2V group was given estradiol valerate(0.6 mg/kg).The ICA-L group,ICA-M group and ICA-H group were given low,medium and high doses of ICA(50,100 and 200 mg/kg)respectively.After the experiment,the femoral bone microstructure,bone mineral density(BMD),relative bone volume(BV/TV),trabecular number(Tb.N)and trabecular separation(Tb.Sp)were measured.Enzyme linked immunosorbent assay(ELISA)was used to detect serum osteoprotegerin(OPG),receptor activator of nuclear factor-KB ligand(RANKL)and tartrate-resistant acid phosphatase 5b(TRACP-5b).Serum levels of superoxide dismutase(SOD),malondialdehyde(MDA)and Ferrous ion(Fe2+)were detected by the kit.The expression levels of ferritin heavy chain 1(FTH1),SLC7A11 and GPX4 in rat lumbar spine were detected by Western blotting.Quantitative real-time reverse transcription polymerase chain reactio(qRT-PCR)was used to detect the expression levels of SLC7A11 mRNA and GPX4 mRNA in rat lumbar tissues.Results:In OVX group,the femur of rats showed significant bone microstructure degradation,which was manifested by the decrease of trabecular number,accompanied by the disorder of trabecular network arrangement,characteristic trabecular structure atrophy and irregular morphological changes.The trabecular density of ICA-L group was slightly improved compared with OVX group.The trabecular structure of femur in E2V group,ICA-M group and ICA-H group was arranged in order.BMD,BV/TV and Tb.N in OVX group were lower than those in Sham group(P<0.01),while Tb.Sp was higher than that in Sham group(P<0.01).The E2V group,ICA-M group and ICA-H group showed higher BMD,BV/TV and Tb.N than OVX group(P<0.05 or P<0.01),while lower Tb.Sp than OVX group(P<0.01).The BMD in ICA-L group was higher than that in OVX group(P<0.05).The contents of OPG,OPG/RANKL and SOD in serum of OVX group were lower than those in Sham group(P<0.01),while the contents of RANKL,TRACP5b,MDA and Fe2+were higher than those in Sham group(P<0.01).The contents of OPG,OPG/RANKL and SOD in serum of E2V group were higher than those of OVX group(P<0.05 or P<0.01),while the contents of RANKL,TRACP5b,MDA and Fe2+were lower than those of OVX group(P<0.01).The content of OPG in serum of ICA-H group was higher than that in OVX group(P<0.01).Serum SOD content and OPG/RANKL in ICA-L group,ICA-M group and ICA-H group were higher than those in OVX group(P<0.01),while the contents of RANKL,TRACP5b,MDA and Fe2+were lower than those in OVX group(P<0.01).The relative expression of FTH1,SLC7A11 and GPX4 protein in OVX group was lower than that in Sham group(P<0.01).The relative expressions of FTH1,SLC7A11 and GPX4 proteins in lumbar vertebrae of E2V group and ICA-H group were higher than those in OVX group(P<0.01).The relative expression of SLC7A11 and GPX4 protein in lumbar vertebrae of rats in ICA-M group was higher than that in OVX group(P<0.01).The relative expression of GPX4 protein in the lumbar vertebrae of rats in ICA-L group was higher than that in OVX group(P<0.05).The relative expressions of SLC7A11 mRNA and GPX4 mRNA in the lumbar vertebrae of OVX group were lower than those in Sham group(P<0.01).The relative expressions of SLC7A11 mRNA and GPX4 mRNA in lumbar vertebrae of E2V group,ICA-L group,ICA-M group and ICA-H group were higher than those in OVX group(P<0.01).Conclusion:Icariin can decrease osteoclast activity in OVX rats,inhibit bone resorption,increase antioxidant capacity,reduce lipid peroxidation damage,reduce free Fe2+accumulation,ultimately reduce bone resorption,promote bone formation,repair trabecular structure and play an anti-osteoporosis role through the up-regulation mechanism of SLC7A11/GPX4 axis.
