Neuroprotective Effects of Minor Xuming Decoction Against Cerebral Ischemia-Reperfusion Injury via Regulation of PINK1/Parkin-Mediated Mitophagy

目的:探讨小续命汤对脑缺血再灌注损伤大鼠的神经保护作用及其作用机制.方法:将 60 只 SPF 级健康雄性 SD 大鼠随机分为假手术组、模型组、小续命汤低剂量组(15 g·kg-1)和小续命汤高剂量组(30 g·kg-1).除假手术组外,其余组大鼠采用改良的Zea-longa 法诱导局灶性脑缺血再灌注模型.模型建立完成后对各组大鼠进行灌胃给药,假手术组和模型组给予等体积的生理盐水,每日1 次,连续7 d.采用 Zea-longa 法对大鼠进行神经功能缺损评分;Morris 水迷宫测试评估大鼠的认知功能;TTC 染色计算大鼠脑梗死体积;透射电镜观察线粒体超微结构;分离线粒体并检测三磷酸腺苷(adenosine triphosphate,ATP)的含量;Western Blot 检测 LC3、Parkin、P62、Beclin-1 蛋白表达水平.结果:与假手术组比较,模型组神经功能评分显著升高,脑梗死体积显著增大;逃避潜伏期显著延长,穿越平台次数显著减少,目标象限停留时间显著缩短;皮层组织 LC3Ⅱ/Ⅰ比值、Beclin-1、Parkin 及 p62 蛋白表达水平显著升高;线粒体数量显著降低,ATP 含量显著下降,差异均具有统计学意义(P<0.05).与模型组比较,小续命汤高剂量组神经功能评分显著降低,脑梗死体积显著减小;逃避潜伏期显著缩短,穿越平台次数显著增加,目标象限停留时间显著延长,游泳速度明显提高;LC3Ⅱ/Ⅰ比值及 Beclin-1、Parkin、p62 蛋白表达水平显著下调;线粒体数量显著增多,ATP 含量显著升高,差异均具有统计学意义(P<0.05).透射电镜显示:模型组神经元线粒体肿胀、嵴断裂、空泡化明显,而小续命汤高剂量组线粒体结构损伤显著减轻.结论:小续命汤对脑缺血再灌注损伤大鼠具有神经保护作用,其机制可能与抑制 PINK1/Parkin 通路的过度激活,调节线粒体自噬,维持线粒体结构与功能完整性有关.

Objective:To investigate the neuroprotective effects of Minor Xuming Decoction on cerebral ischemia-reperfusion injury(CI-RI)and its underlying mechanisms.Methods:A total of 60 healthy male SPF-grade SD rats were randomly divided into four groups:sham-operated group,model group,Minor Xuming Decoction low-dose group(15 g·kg-1)and Minor Xuming Decoction high-dose group(30 g·kg-1).Except the sham-operated group,rats in all groups were established focal cerebral ischemia-reperfusion injury models using the modified Zea-Longa method.After model establishment,rats in each group were given intragastric administration of cor-responding drugs once daily for 7 consecutive days;rats in the sham-operated and model groups received an equal volume of normal sa-line instead.The Zea-Longa neurological deficit scale was adopted to score neurological dysfunction.The Morris water maze test was per-formed to evaluate cognitive function.TTC staining was used to calculate cerebral infarct volume.Transmission electron microscopy(TEM)was applied to observe mitochondrial ultrastructure.Mitochondria were isolated to detect adenosine triphosphate(ATP)content.Western blotting was utilized to measure the protein expression levels of LC3,Parkin,p62 and Beclin-1.Results:Compared with the sham-operated group,the model group showed remarkably higher neurological deficit scores and larger cerebral infarct volumes.In the Morris water maze test,rats in the model group presented prolonged escape latency,fewer platform crossings,and shorter residence time in the target quadrant.For cortical tissue protein detection,the LC3-Ⅱ/LC3-Ⅰ ratio,as well as the expression levels of Beclin-1,Parkin and p62,were significantly upregulated.In addition,mitochondrial number and ATP content were markedly reduced.All above differ-ences were statistically significant(P<0.05).Compared with the model group,the Minor Xuming Decoction high-dose group exhibited significantly lower neurological deficit scores and smaller cerebral infarct volumes.Meanwhile,the rats presented shortened escape laten-cy,increased number of platform crossings,prolonged residence time in the target quadrant,and markedly elevated swimming speed.In addition,the LC3-Ⅱ/LC3-Ⅰ ratio and the expression levels of Beclin-1,Parkin and p62 were significantly downregulated,while mito-chondrial number and ATP content were remarkably increased.All differences above were statistically significant(P<0.05).Conclu-sion:Minor Xuming Decoction exerts neuroprotective effects against CIRI.Its underlying mechanism may be associated with inhibiting excessive activation of the PINK1/Parkin pathway,modulating mitophagy,and preserving the structural and functional integrity of mito-chondria.
