抗多花黄精根腐病生防菌A3-1的促生防病机理OA
Plant Growth-Promoting and Disease-Preventing Mechanisms of Biocontrol Strain A3-1 Against Root Rot of Polygonatum cyrtonema Hua
为探究多花黄精(Polygonatum cyrto-nema Hua)根腐病的绿色生防方法,缓解新化黄精连作障碍,以病株上分离的 4 种病原菌为靶标,以实验室筛选鉴定的枯草芽孢杆菌(Bacillus subtilis)A3-1 为生防菌,通过平板对峙试验、促生与抑菌功能验证、代谢产物抑菌活性评估及菌丝形态观察,系统评价了 A3-1 对黄精根腐病的生防潜力.结果表明:A3-1 具有产氨、固氮、分泌嗜铁素、溶解有机磷与无机磷等多种促生特性,其ACC 脱氨酶活性达 65.04 U/mL,并能分泌纤维素酶、蛋白酶和淀粉酶;平板对峙试验中,该菌株对白蜡树炭疽菌(Colletotrichum spaethianum)抑制效果最佳,而其挥发性代谢产物对腐皮镰刀菌(Fusarium solani)的抑制率高达91.39%,同时引发了菌丝形态畸变,但其无菌滤液未表现出明显抑菌活性.综上,菌株 A3-1 兼具促生与抑菌双重功能,其挥发性气体为关键抑菌因子,具有良好的生防应用前景.
This study aimed to explore the green biocontrol methods for root rot of Polygonatum cyrtonema Hua and alleviate the continuous cropping obstacles of P.cyrtonema in Xinhua County,Hunan Province.Four pathogenic fungal species isolated from diseased plants were taken as the target pathogens,and Bacillus subtilis A3-1 screened and identified in our laboratory was used as the biocontrol agent.The biocontrol potential of A3-1 against root rot of P.cyrtonema was systematically evaluated through plate confrontation experiments,evaluation of plant growth-promoting and fungal pathogen-inhibiting functions,antifungal activity evaluation of metabolites,and hyphal morphology observation.The results showed that A3-1 had multiple plant growth-promoting functions such as producing ammonia,fixing nitrogen,secreting siderophores,and solubilzing organic and inorganic phosphorus.Its ACC deaminase activity reached 65.04 U/mL,and it could secrete cellulase,protease,and amylase.The plate confrontation experiments showed that the strain exhibited the best inhibitory effect on Colletotrichum spaethianum,while its volatile metabolites had a high inhibition rate of 91.39%on Fusarium solani,while causing morphological distortion of hyphae.However,the sterile filtrate of this strain did not show significant antifungal activity.In summary,strain A3-1 showed dual functions of promoting plant growth and inhibiting fungal pathogens,with its volatile gas as the key antifungal component,demonstrating promising application prospects in biocontrol.
