多基因聚合培育抗稻瘟病且镉低积累水稻新种质OA
Breeding of New Rice Germplasm with Rice Blast Resistance and Low Cadmium Accumulation by Gene Pyramiding
为了培育抗稻瘟病且籽粒镉低积累的水稻新种质,以携带稻瘟病抗性基因 Pita的水稻两系不育系知 796S、保持系金珍 B、携带Pi2基因的恢复系职香恢859及聚合稻瘟病抗性基因的恢复系中间材料 KZ-3425 为受体亲本,以携带 OsNramp5 缺失基因型的珞红 4B 和湘安 1 号为供体亲本,采用分子标记辅助选择技术,通过杂交、回交及多代自交选育,将稻瘟病抗性基因(Pita/Pi2)与镉低积累基因 OsNramp5 缺失等位基因聚合到同一遗传背景中.结果表明:聚合Pi2基因的改良株系稻瘟病抗性达到中抗(3级)水平,较受体亲本(中感 5 级)显著提升;所有改良株系糙米镉含量范围为 0.011~0.035 mg/kg,远低于国家食品安全标准限量值(0.2 mg/kg).农艺性状调查显示,不同遗传背景改良株系的农艺性状存在明显差异,其中以职香恢859为遗传背景的 LF104、LF106、LF107 等株系综合性状优良,兼具中抗稻瘟病和籽粒镉低积累特性,可为镉污染稻区的绿色安全生产提供材料基础和理论参考.
This study aimed to breed new rice germplasm with rice blast resistance and low cadmium accumulation in grains.The two-line sterile rice line Zhi 796S carrying the rice blast resistance gene Pita,the maintainer line Jinzhen B,the restorer line Zhixianghui 859 carrying Pi2,and the intermediate material KZ-3425 of the restorer line that carried the rice blast resistance gene were used as receptor parents.Luohong 4B and Xiang'an 1 with OsNramp5 deletion were used as the donor parents.Molecular marker-assisted selection was used to integrate the rice blast resistance gene(Pita/Pi2)and the allele of low cadmium accumulation gene OsNramp5 deletion into the same genetic background through crossing,back-crossing,and multi-generation selfing.The results showed that the obtained rice line carrying Pi2 exhibited moderate resistance(grade 3)to rice blast,which was significantly higher than that(moderate susceptibility,grade 5)of the receptor parent.The obtained lines showed the cadmium content within the range of 0.011-0.035 mg/kg,which was far below the limit of 0.2 mg/kg specified in the national food safety standard.There were significant differences in the agronomic traits of improved lines with different genetic backgrounds.The lines LF104,LF106,and LF107 with the genetic background of Zhixianghui 859 demonstrated excellent comprehensive traits and possessed moderate resistance to rice blast and low cadmium accumulation in grains.This study provided a material basis and theoretical reference for green and safe rice production in cadmium-contaminated areas.
