水飞蓟素通过增强抗氧化能力改善湖羊精液冷冻保存效果的研究OA
Study on silymarin improving the cryopreservation effect of Hu sheep semen by enhancing antioxidant capacity
为探究水飞蓟素(SL)对湖羊精液冷冻保存效果的影响,试验选取 3 只 2~3 岁、体重相近、体况良好的湖羊为研究对象,分别隔天采精 3 次,将检测合格的精液混合后用含不同浓度 SL 的冷冻稀释液稀释后冷冻保存,对照组不添加 SL,试验组分别添加 12.5,25,50,100,200 μmol/L 的 SL,解冻后检测精子活率(TM)、活力(PM)、运动性能[直线速度(VSL)、曲线速度(VCL)、路径速度(VAP)和鞭打频率(BCF)]、质膜完整率、顶体完整率、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)水平、线粒体膜电位(MMP)及活性氧(ROS)水平.结果表明:与对照组相比,50 μmol/L 组精子活率、活力、运动性能、质膜完整率、顶体完整率、线粒体膜电位水平、CAT 和 SOD 活性显著提高(P<0.05),MDA 和 ROS 水平显著降低(P<0.05).说明 50 μmol/L SL 可通过增强精子抗氧化能力以减轻冷冻损伤.
In order to explore the effects of silymarin(SL)on the cryopreservation of semen of Hu sheep,three 2-3 year old Hu sheep with similar body weight and good body condition were selected as the research objects,and the semen was collected three times every other day.The qualified semen was mixed and diluted with the frozen diluent containing different concentrations of SL.In the control group,no SL was added to the semen,while in the experimental groups,12.5,25,50,100,and 200 μmol/L of SL were added,respectively.After thawing,sperm motility(TM),motility(PM),motility performance(linear velocity[VSL],curvilinear velocity[VCL],path velocity[VAP]and beat frequency[BCF]),plasma membrane integrity rate,acrosome integrity rate,malondialdehyde(MDA)content,superoxide dismutase(SOD)activity,catalase(CAT)activity,mitochondrial membrane potential(MMP)and reactive oxygen species(ROS)level were measured.The results showed that compared with the control group,the sperm motility,motility,exercise performance,plasma membrane integrity rate,acrosome integrity rate,mitochondrial membrane potential level,activities of CAT and SOD(P<0.05)were significantly increased in the 50 μmol/L group,while the levels of MDA and ROS(P<0.05)were significantly decreased.The results indicated that 50 μmol/L SL could reduce freezing damage by enhancing the antioxidant capacity of sperm.
